摘  要:目的系统研究牛膝中齐墩果酸皂苷及其富集工艺。方法采用UV方法确定牛膝中齐墩果酸的最佳的提取方法,利用大孔树脂研究了齐墩果酸皂苷的吸附与解吸附作用,优化了大孔树脂富集纯化齐墩果酸皂苷的工艺。利用比较合适的齐墩果酸-28-葡萄糖苷为标准品,建立了紫外显色法测定齐墩果酸总皂苷含量的方法。结果牛膝中齐墩果酸皂苷的最佳提取条件为70%乙醇提取回流2h。利用HP-20大孔树脂处理以及采用新的标准品为对照,提取物中总皂苷含量可提高到64.65%。结论利用齐墩果酸-28-葡萄糖苷为标准品,HP-20富集、纯化牛膝总皂苷是可行的。OBJECTIVE To systematically study the oleanolic acid type saponins and its enrichment process in roots of Achyranthes bidentata. METHODS With UV method, extraction method of the saponins in the roots was established. Using macroporous resins, the adsorptive and desorptive effects on the saponins were evaluated, and the enrichment process of the saponins was optimized. With oleanolic acid-28-O-glucoside as a new standard, a UV method on the quantification of the total oleanolic acid type saponins was established. RESULTS The optimized extract method was 70 % ethanol and reflux 2 hours. Treating with HP-20 macroporous resins and using a new standard, the content of the total saponins in the processed extract reached to 64.65%. CONCLUSION It is feasible to enrich and purify the saponins of Achyranthes bidentata with HP-20 and oleanolic acid 28-O-glucoside as a new standard.
