摘 要:目的评价甘遂不同提取物对模式生物斑马鱼的急性毒性。方法采用回流提取方法制备甘遂水提物、醇提物、先醇提后水提物;将甘遂不同提取物按几何级数设置浓度梯度,添加到鱼只生活的水中,观察给药后96h鱼只的死亡情况,以此为判断待测药物毒性大小的依据,采用SPSS 13.0软件计算各药物对斑马鱼的半数致死浓度(LC50)。结果斑马鱼对甘遂不同提取物均表现出急性毒性反应,并呈现出明显的量-毒关系;甘遂水提物LC50为31.00μg/mL,甘遂醇提物LC50为6.89μg/mL,甘遂先醇提后水提物LC50为4.26μg/mL。结论以斑马鱼作为实验动物,发现甘遂先醇提后水提物急性毒性最强,水提物毒性最弱,为进一步认识与评价甘遂的毒性作用提供了依据。OBJECTIVE To evaluate the acute toxicity of different kansui extracts on the model organism zebrafish. METHODS The kansui water extract, alcohol extract, water-alcohol extract were prepared by reflux extraction method; The death of zebrafish was observed after explosion for 96 h to different kansui extracts classified into different degrees, which was the judge for drug toxin basis. The half lethal concentrations (LC50) of different kansui extracts on zebrafish were calculated by SPSS13.0 software. RESULTS Zebrafish in different extracts all represented acute toxicity reaction and shoed apparent dosetoxicity relationship; the LC50 values of the water extract, the alcohol extract and the water-alcohol extract were 31.00μg/mL, 6.89 μg/mL, and 4.26μg/mL, respectively. CONCLUSION With zebrafish as experimental animals, the water-alcohol extract of kansui possesses the highest toxicity, whereas the water extract is lowest. The result sprovide evidence for the further understanding and evaluation on the toxicity of kansui.
