摘  要:目的:观察地尔硫芯卓对实验家兔动脉粥样硬化模型的干预作用,并探讨钙拮抗剂抗动脉粥样硬化的作用机制。方法:36只大白兔随机分为4组,正常组对照组、高脂对照组、阿托伐他汀组、地尔硫芯卓组,每组9只。观察实验后家兔血总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL)、白细胞介素6(IL-6)、C反应蛋白(CRP)在各组间的含量变化,测定实验结束时家兔主动脉粥样硬化斑块的面积及显微镜下病理形态表现。结果:与高脂组相比,阿托伐他汀组TC、TG、LDL-C、IL-6、CRP含量明显降低(P<0.01),主动脉粥样硬化面积明显降低(P<0.01);与高脂组相比,地尔硫芯卓组TC、LDL-C、IL-6、CRP降低(P<0.01),主动脉粥样硬化面积明显降低(P<0.01);与阿托伐他汀组相比,地尔硫芯卓组IL-6明显降低(P<0.01),CRP、TC、TG明显升高(P<0.01)。结论:钙拮抗剂地尔硫芯卓对动脉粥样硬化有明显的预防和治疗作用,其机制可能和抗氧化、减少炎性介质IL-6、CRP的释放、改善内皮细胞功能有关。OBJECTIVE To investigate the mechanism of diltiazem against the formation of atherosclerosis in rabbits, and to elucidate the anti-atherosclerotic mechanism of calcium channel blocker(CCB). METHODS 36 healthy male rabbits were randomly divided into four groups: controlled group (9 cases) ; high-fat diet group (9 cases) ; atorvastatin group (9 cases) ; diltiazem group (9 cases). After twelve weeks experiment each group's triacylglyceral, total cholesterol, low density cholesterol, high density lipoprotein cholesterol, C-reactive protein and interleukin-6 were measured. The area of athcrosclcrosis mottle in tunica intima of aorta(AAMA) was computed. RESULTS Compared with the high-fat diet group, the atorvastatin groups TC. TG. LDL-C. IL-6, CRP and AAMA significantly lowered(P〈0.01 ) ; Compared with the high-fat diet group , the diltiazem "groups TC.LDL-C, IL-6,CRP and AAMA significantly lowered(P〈0. 01 ); Compared with atorvastatin group, the diltiazem' groups IL-6 significantly lowered(P〈0. 01) ,TG,TC,CRP remarkably increased(P〈0. 01). CONCLUSION Diltiazem can reduce the releass of inflammatory mediators IL-6 and CRP effectively against athcrosclcrosis.
