摘  要:品种鉴定是种质资源研究和新品种保护的基础,研究在形态学鉴定的基础上,通过ISSR和SSR标记,对3株北京28号疑似芽变材料及来自同一果园和不同果园对照未变异北京28号进行分子鉴定。结果发现,芽变株系在物候期及果实性状上存在不同程度的变异,50对SSR标记中只有CPPCT022和BPPCT023在对照C2和其他供试样品间存在不同等位基因位点;39条ISSR引物共扩增出314条DNA片段,其中36条具有多态性,多态性频率为11.46%;3株疑似芽变材料两两之间遗传相似系数达到0.99,大于来自同一果园的两对照C1和C2间的遗传相似系数(0.96),说明3株疑似芽变材料应为同一芽变枝嫁接而来。通过UPGMA聚类分析发现,当遗传相似系数为0.96时,3株疑似芽变材料与来自同一果园的两对照未变异北京28号聚为一类群,且与对照C2亲缘关系最为密切,结合表型分析,说明3株疑似芽变株系很可能是由对照C2芽变而来,而CPPCT022和BPPCT023有可能是其变异位点。Molecular identification is important for the protection of new varieties and studies of germplasm resources. Of fingerprinting techniques, Inter-Simple Sequence Repeat (ISSR) and Simple Sequence Re- peat (SSR) were among the most informative and powerful. In this study, we identified a peach mutation cuhivar from Beijing 28 by ISSR and SSR based on the identification of phenotypic characters. The results showed that different variations of phenophase and fruit characters were observed in bud sports. The SSR primers could produce one or two discrete fragments in these accessions, and polymorphic bands were found between C2 and other samples in CPPCT022 and BPPCT023. Thirty nine ISSR primers amplified 314 bands, 36 bands were polymorphic, and the polymorphic percentage was 11.46%. The genetic simi- larity coefficient (GS) estimated from these molecular data ranged from 0.91 to 0.99, with an average of 0.95. The GS of three mutation accessions was 0.99 and of two controls from same orchard was 0.96. The combined grouping association indicated that these mutation accessions and two controls from the same or- chard were clustered distinctly, and the genetic relationship was most closely between C2 and mutations. The three bud sports should be most likely from the control C2 combined the identification of phenotypic variation, and CPPCT022 and BPPCT023 might be the variation sites. These results provided valuable references for identification and protection of a new variety, classification and application of peach germplasm in commercial nurseries.
