摘 要:[目的]采用超声技术提取秦艽中主要成分龙胆苦苷,建立高效液相色谱技术测定龙胆苦苷含量的方法。[方法]采用单因素试验,考察超声频率、超声功率、提取时间、提取剂用量对超声波提取的影响,以正交试验确定最佳提取条件;采用高效液相色谱法进行含量测定。[结果]最佳工艺条件为超声频率为80 KHz,超声功率为420 W,提取时间为20 min,提取剂用量为20 ml;在此情况下,龙胆苦苷的提取率为2.72%。[结论]该方法灵敏、准确、简便,适用于秦艽中龙胆苦苷的提取和含量测定。[ Objective] The aim was to extract gentiopicroside from G. macrophylla by using ultrasonic technology and the method for content determination of gentiopicroside by HPLC was set up. [ Method ] Orthogonal experiments were carried out to investigate the effect of ultrasonic frequency, ultrasonic power, ultrasonic time and extractant amount on ultrasonic extraction, and the best condition was determined. Contents of gentiopieroside were determined by HPLC. [ Result] The best extraction conditions of gentiopicroside were as below: ultrasonic frequency was 80 KHz, ultrasonic power was 420 W, ultrasonic time was 20 min, and extractant amount was 20 ml. [ Conclusion ] This method was sen- sitive, accurate, simple and could be used in the determination of gentiopicroside in G. macrophylla.
