摘 要:目的:探讨雌激素对人乳腺癌细胞周期蛋白E表达及在血清饥饿状态下细胞存活力的影响。方法:以MCF-7乳腺癌细胞为模型,采用蛋白印迹分析法检测乳腺癌周期蛋白与表达、显微镜下观察细胞变化、台盼蓝染色及细胞计数法测算细胞存活力。结果:雌激素(10 nmol/L)处理可引起乳腺癌细胞周期蛋白E的过度表达,与对照组比较,增高56.42%(P<0.05);在无血清培养条件下,雌激素能减少乳腺癌细胞死亡,增强细胞的存活力,与对照组比较,72 h细胞存活率增高19.86%(P<0.05)。结论:雌激素能显著上调周期蛋白E的蛋白表达,并增强乳腺癌细胞对血清饥饿的抵抗力。Objective: To explore the effects of estrogen on the expression of cyclin E and cell viability during serum starvation in human breast cancer cells.Methods: MCF-7 human breast cancer cells were employed as study model.Western blotting was used to detect the expression of cyclin E protein.Changes of cell morphology were observed under microscope.Trypan Blue staining and cell counting were employed to determine cell viability.Results: In breast cancer cells,estrogen(10 nmol/L) could increase the expression of cyclin E by 56.42% as compared with that of control(P0.02).On the condition of serum-free culture,estrogen reduced cell death and increased cell viability.With the action of estrogen,cell viability increased gradually with the time passing.Cell viability increased by 19.86% in 72 h as compared with that of control(P0.05).Conclusions: Estrogen contributes to increased expression of cyclin E and enhances resistance of breast cancer cells to serum starvation.
