摘 要:为了检测MyoG基因和A-FABP基因单核苷酸多态性(SNP)与鹅肉品质性状的相关性,以太湖鹅为研究对象,测定肌内脂肪含量和相关组织学指标特性,利用PCR-SSCP和测序技术研究MyoG基因外显子1和A-FABP基因内含子2SNPs,并进行相关分析。研究结果表明:(1)MyoG基因外显子1扩增序列大小为420bp,共有1个突变,突变位点在64bp处,为C→T突变,不同基因型之间肌纤维密度、直径差异显著(P<0.05);(2)A-FABP基因扩增序列大小为482bp,共发现3个突变位点,在第187bp处发生C→T突变,在第235bp处发生C→A突变,在第372bp处发生A→C突变;(3)A-FABP基因内含子2扩增产物经SSCP分析后发现两种基因型,不同基因型之间脂肪含量有一定差异。比对发现其与家鸭同源性达91%,与家鸡的同源性为85%。本研究结果为进一步分析MyoG和A-FABP基因的遗传变异及其与肉质性状的相关性以及将该分子标记应用于育种计划奠定一定的理论基础。In order to detect the relativity of single nucleotide polymorphism of MyoG gene and adipo- cyte fatty acid binding protein (A-FABP) gene with meat quality in the geese, the crude fat and some muscle fiber traits were detected, using the techniques of PCR-SSCP and sequencing to study the single nucleotide polymorphism of exon 1 of MyoG gene and intron 2 of A-FABP of Taihu geese, and then relativity analysis was conducted. The results showed: (1) The PCR products of exon 1 of MyoG gene had about 420 bp, the mutational site was on the 64 bp (C→T). There was a significant difference of muscle fiber density and muscle fiber diameter between different genotypes(P〈0.05). (2) The products of PCR had about 482 bp, the intron 2 of A-FABP was from 174 bp to 394 bp, there were three mutational sites, respectively on the 187 bp (C→T), 235 bp (C→A) and 372 bp(A→C) ; (3) By using PCR-SSCP ,two genotypes of the intron 2 of A-FABP were found, there had some difference between different genotypes. Compared the sequence of intron 2 of goose with duck and chicken, the homology were 91% and 85%, respectively. These findings laid good theoretical foundation for the progressive research of the relationship between genetic mutation in MyoG and A-FABP gene with meat trait and for the progressive research of the application of molecular markers in animal breeding.
