摘 要:目的观察络通纤溶饮对盐酸博莱霉素诱导特发性肺纤维化(IPF)大鼠肺组织中羟脯氨酸(HYP)含量的影响。方法将SD大鼠120只随机分为正常组、模型组、络通纤溶饮低剂量组、络通纤溶饮高剂量组、泼尼松组5组,每组24只(每组再随机分3批,每批8只)。正常组气管内滴注0.9%氯化钠注射液,其余4组大鼠气管内滴注盐酸博莱霉素溶液造模。造模后正常组、模型组分别按照10 mL/kg体质量予0.9%氯化钠注射液灌服,每日1次;络通纤溶饮低剂量组和高剂量组分别按照12.3、24.6 g/kg体质量予络通纤溶饮水煎剂灌服,每日1次;泼尼松组按照6 mg/kg体质量予泼尼松混悬液灌服,每日1次。各组大鼠分别于给药后第7、14、28 d断头处死,取右肺中叶组织检测HYP的含量。结果 28 d内正常组大鼠无死亡,模型组死亡6只,络通纤溶饮低剂量组死亡3只,络通纤溶饮高剂量组死亡3只,泼尼松组死亡4只。与本组前一时间段及正常组比较,其余4组HYP含量持续增高(P<0.05);与模型组比较,同一时间段络通纤溶饮高剂量组、络通纤溶饮低剂量组和泼尼松组HYP含量均降低(P<0.05);与泼尼松组比较,络通纤溶饮高剂量组、络通纤溶饮低剂量组HYP均降低,给药第14、28 d络通纤溶饮高剂量组HYP含量与泼尼松组比较差异有统计学意义(P<0.05);络通纤溶饮高剂量组、低剂量组同一时间段HYP含量比较差异无统计学意义(P>0.05)。结论络通纤溶饮可抑制造模状态下IPF大鼠肺组织中HYP含量的异常增高。Objective To investigate the effect of Luotongqianrongyin on the eontent of hydmxyproline (HYP) in rat lung tissue with idiopathic pulmonary fibrosis (IPF) induced by bleomycin HCI. Methods 120 SD rats were randomly divided into five groups, control group, model group, Luotongqianmngyin low dose group, Luo- tongqianmngyin high dose group and Prednisone group, n = 24 (each group were randomly divided into three bat- ches, each hatch of eight). Normal group received intratraeheal instillation of 0.9% sodium ehloride injection, and the remaining 4 groups reeeivod intratracheal instillation of bleomyein solution for modeling of IPF. After modeling control group and model group received 0.9% sodium chloride injeetion,10mL/kg. Luotongqianrongyin low dose group and Luotongqianmngyin high dose group received Luotongqianmngyin, 12.3 and 24.6 g/kg, once a day re- spectively. Prodnisone group received by 6 mg/kg body weight to the prednisone suspension fed, once a day. The levels of HYP in right middle lobe tissue were detected after the administration of 7, 14, 28 d rats were decapitated. Results The levels of HYP on 28 days in model group, Luotongqianmngyin low dose group, Luotongqianmngyin high dose group and Prednisone group were increased in comparison with those control group and these four groups of previous time period ( P 〈 0.05 ). The levels of HYP in Luotongqianrongyin low dose group, Luotongqianrongyin high dose group and Prednisone group were decreased as compared with those in model group at same time period(P 〈 0.05). The levels of HYP in Luotongqianrongyin low dose group and Luotongqianrongyin high dose group were de- creased as compared with those in Prednisone group. There was significant difference between Luotongqianrongyin high dose group and Prednisone group on the level of HYP at ad-ministration of 14, 28d ( P 〈0.05). Conclusion Luotongqianrongyin can inhibit the content of abnormally high levels of HYP in IPF lung tissue in the state of modeling.
