摘 要:目的:建立大鼠血浆中葡萄内酯的检测方法,测定葡萄内酯与大鼠血浆蛋白的结合率。方法:采用平衡透析法研究葡萄内酯在大鼠体内与血浆蛋白结合率。利用醋酸乙酯萃取富集袋内外样品中的葡萄内酯,并用HPLC法测定其浓度,进而求出血浆蛋白结合率。结果:在低、中、高3种浓度下,葡萄内酯的血浆蛋白结合率分别为100.0%、(99.7±0.6)%、(99.1±0.8)%。结论:葡萄内酯溶液与大鼠血浆蛋白属高度结合。OBJECTIVE To establish a chromatographic method of auraptene in rats plasma, and determine the binding rate of auraptene with rats plasma protein. METHODS The binding rate of auraptene with rats plasma was investigated with equilibrum dialysis method. Auraptene in the dialysate was concentrated with acetic ether, and its contents were determined by HPLC, then the binding rates were calculated. RESULTS The binding rate of auraptene with rats plasma protein was 100. 0%, (99. 7±0. 6) %, and (99. 1± 0. 8)%, respectively. CONCLUSION The binding rates of auraptene with rat plasma protein belong to the high-rate combination.
