摘 要:目的探讨中华眼镜蛇毒细胞毒素-12(CTX-12)联合射线处理,对Balb/c裸鼠皮下移植膀胱肿瘤的生长抑制作用。方法建立40只Balb/c雌性裸鼠皮下移植瘤的动物模型,待肿瘤体积达到(0.60±0.09)cm3时,随机将动物按处理方法的不同分成CTX-12+放射组、单纯CTX-12组、单纯放射组和空白对照组,观察并比较各组的抑瘤率,并以流式细胞术、TUNEL系统检测肿瘤细胞凋亡情况。结果单纯放射组、单纯CTX-12组、CTX-12+放射组的抑瘤率分别为18.2%、46.7%、77.9%;与对照组比较,三组动物的皮下移植肿瘤都受到抑制(P<0.05);但CTX-12+放射组肿瘤受抑制程度明显高于单纯CTX-12组(P<0.05)和单纯放疗组(P<0.01);肿瘤细胞凋亡率及凋亡指数亦然。结论 CTX-12联合射线处理能显著抑制Balb/c裸小鼠皮下移植膀胱肿瘤的生长。CTX-12联合生理剂量射线主要是通过诱导肿瘤细胞凋亡而达到明显抗膀胱肿瘤作用。Objective To investigate the antitumor efficacy of cytotoxin 12(CTX-12) from cobra (Naja naja atra) venom in combination of radiotherapy on Balb/c nude mice transplanted with hypodermal bladder tumor.Methods The hypodermal transplantation tumor models of 40 Balb/c female nude mice (6 to 8 weeks of age) were established.When transplantation bladder tumor turned to be (0.60±0.09) cubic centimeter in volume,40 nude mice were randomly divided into 4 groups (n=10):CTX-12 plus radiotherapy,CTX-12 alone,radiotherapy alone and control group.The tumor weight,tumor volume inhibitory ratio,flow cytometry and TUNEL systerm for apoptotic circumstances were used to evaluate the treatment effects of three different means of therapy.Results The tumor inhibition rates from radiotherapy alone,CTX-12 alone and CTX-12 with radiotherapy group were 18.2%,46.7% and 77.9%,respectively,which are all significantly higher in inhibitive effect compared with control group (P0.05).In addition,the CTX-12 with radiotherapy group shows more significant tumor suppressive effect than CTX-12 alone (P0.05) and radiotherapy alone group (P0.01).The results are similar in the apoptotic ratio and apoptotic index investigation.Conclusions Cytotoxin-12(CTX-12) from cobra (Naja naja atra) venom in combination of radiotherapy can inhibit conspicuously the growth of hypodermal transplantation bladder tumor,which achieve antitumor effect by inducing tumor cell apoptosis.
