不同来源黄芪多糖提取物对淋巴细胞增殖的影响及其化学组成分析

摘　　要：目的考察不同来源黄芪多糖提取物对体外脾淋巴细胞增殖的影响,明确其活性差异,对比分析其化学组成。方法 MTT法分别检测黄芪多糖提取物对正常脾淋巴细胞、ConA活化脾淋巴细胞增殖的影响;硫酸苯酚法结合重量法分别测定样品中总糖、多糖、醇不溶组分含量。结果各批样品对正常脾淋巴细胞均有显著的促增殖作用(P<0.01或P<0.05),除SSK外,各样品在低浓度时促增殖作用较强,促增殖率为39.4%～186.4%;XZX、XHS、SLH1对ConA诱导的脾淋巴细胞增殖具有显著抑制作用(P<0.01),且在4 mg.ml-1时抑制作用最强,抑制率为52.9%～104.0%,作用大小为XZX>XHS>SLH1,其余各批样品无明显抑制作用;各批样品醇不溶组分含量为18.74%～60.67%,多糖含量为7.22%～44.74%,大小顺序为XZX>XHS>SLH1>SLH2>SSK。结论不同来源黄芪多糖提取物的质量差异十分显著。Objective To investigate the effect of Astragalus polysaccharides extract from different sources on lymphocyte proliferation,illuminate the difference of immunocompetence,and compare their chemical composition. Methods MTT method was used to detect the effect of lymphocyte proliferation.Gravimetric analysis was used to determine the content of composition in dissoluble in alcohol,and phenol-sulferic acid method determine the contents of total sugar,polysaccharide in different samples by phenol-sulferic acid method. Results All extracts could remarkably promote normal lymphocyte proliferation（P 0.01 or P 0.05） but SSK,all possessed better effect under lower concentration.The ratio of promoting lymphocyte proliferation fluctuated from 39.4% to 186.4%.XZX,XHS and SLH1 could remarkably inhibit the lymphocyte proliferation activated by ConA（P 0.01）,and had the best effect under 4 g·ml-1,the ratio of restrainning lymphocyte proliferation fluctuated from 52.9 % to 104.0 %,the order in the content magnitude was XZX XHS SLH1,and the rest samples had the same effect.The content of the undissoluble compositions in alcohol fluctuated from 18.74 % to 60.67 %.The polysaccharide content fluctuated from 7.22 % to 44.74%,the order was XZXXHSSLH1SLH2SSK. Conclusion Astragalus polysaccharides from different sources show very remarkable difference in quality.
