摘 要:目的观察眼针对脑缺血再灌注损伤(CIRI)模型大鼠脑组织中MyD88表达,探讨眼针治疗脑损伤的作用机制。方法健康雄性SPF级Wistar雄性大鼠60只,随机分为正常组、假手术组、CIRI模型组和眼针组。CIRI模型组和眼针组采用改良的线栓法建立大鼠大脑中动脉缺血(MACO)再灌注动物模型。RT-PCR及免疫组织化学方法检测大鼠缺血侧脑皮层MyD88的mRNA和蛋白表达的变化。结果与正常组及假手术组比较,模型组大鼠缺血侧脑皮层MyD88 mRNA和蛋白水平均显示出高表达(P<0.01),眼针组同模型组比较,MyD88 mRNA和蛋白表达则下调(P<0.05)。结论眼针抑制脑皮层组织中MyD88的表达,可能是眼针治疗CIRI的主要作用机制之一。Objective To evaluate the expression of MyD88 in cerebral ischemia-reperfusion injury(CIRI) in rat brain cortex and the effect of eye acupuncture therapy. Methods Adult male SPF Wistar male rats were randomly divided into normal group,sham operation group,CIRI model group and eye acupuncture group,15 rats in each group.The model of middle cerebral artery occlusion(MACO) was established by using improved suture method.RT-PCR method and immunohistochemical method were performed to examine the mRNA and protein expression of MyD88 in ischemic brain cortex. Results The mRNA and protein expression of MyD88 in CIRI model group were increased significantly(P0.01) compared with normal group and sham operation group,and which were decreased significantly(P0.05) in eye acupuncture group. Conclusion Eye acupuncture therapy can reduce the expression of MyD88 in CIRI,which may be one of mechanisms of eye acupuncture therapy.
