摘 要:旨在扩增牦牛多杀性巴氏杆菌外膜蛋白OmpH的编码基因,并预测OmpH蛋白二级结构和B细胞抗原表位,从而探讨牦牛多杀性巴氏杆菌外膜蛋白OmpH在免疫保护中所起的作用。对牦牛多杀性巴氏杆菌的外膜蛋白OmpH基因进行PCR扩增及序列测定。应用生物信息学相关软件和方法,对牦牛多杀性巴氏杆菌OmpH蛋白的二级结构和B细胞抗原表位进行预测。牦牛多杀性巴氏杆菌OmpH基因全长1 478 bp,ORF包含1 002 bp编码333个氨基酸。二级结构以无规卷曲为主,有少量的α-螺旋和延伸带;推测OmpH蛋白有1个细胞黏附位点、2个糖基化位点。The aims of this study was to amplify and sequence the OmpH gene of pasteurella multocida,and predict its secondary structure and B-cell epitopes,which provides a theoretical basis for the study of immune mechanism and Pasteurella multocida subunit vaccine.The pasteurella multocida OmpH gene was amplified and sequenced.Using bioinformatices softwares and methods,the secondary structure and B-cell preponderant epitope of pasteurella multocida OmpH were predicted.The results indicated that pasteurella multocida OmpH gene was 1 478 bp,ORF including 1 002 bp,encoding 333 amino acids.Using two methods,prediction of the secondary structure of pasteurella multocida OmpH indicated that random coils were the main structural type of the flexible region in secondary structure,and contain a small amount of α-helix and extended strand.The OmpH protein was supposed contain 1 cell attachment sequence and 2 potential glycosylated sites.These results provide a theoretical basis for the further study of immune mechanisms,monoclonal antibodies preparation and epitope vaccine design.
