卡维地洛对急性心肌梗死大鼠心肌组织中β-arrestin 2蛋白表达的影响

摘　　要：目的:探讨卡维地洛对急性心肌梗死(MI)大鼠非梗死区心肌组织中β-arrestin 2的影响。方法:30只大鼠随机分成假手术组、模型组和卡维地洛组。建立MI模型,术前3d至术后4d给予药物及0.9%氯化钠,实验结束后测量心功能和非梗死区心肌组织中β-arrestin 2的表达。结果:与假手术组相比,模型组大鼠左室收缩末期压(110.72±13.27)mmHg(1mmHg=0.133kPa)、左室压力最大上升速率(2.62±0.83)和下降速率(-2.11±0.57)明显降低(P<0.05)。与模型组比较,卡维地洛对心功能有明显改善作用,可使左室收缩末期压(130.78±7.94)mmHg明显升高(P<0.05)、左室舒张末期压[(12.34±5.55)mmHg]明显降低(P<0.05),并升高了左室压力最大上升速率和下降速率(P<0.05)。模型组大鼠非梗死区心肌β-arrestin 2蛋白表达增加(P<0.05),卡维地洛可进一步增加非梗死区心肌β-arrestin 2蛋白表达(P<0.05)。结论:卡维地洛可明显改善大鼠MI后左室功能,其机制可能与其增加MI后非梗死区心肌组织中β-arrestin 2的表达有关。Objective：To study the effect of carvedilol on cardiac function and the expression of β-arrestin 2 in rats with acute myocardial infarction（MI）.Method：All rats were randomized to the following groups ： sham gro-up,MI group and carvedilol group.All rats were given vehicle or carvedilol by direct gastric gavage for 7 days.Four days after MI,infarct size,ventricular weight/body weight ratio,LV function,non-infarcted myocardial collagen volume fraction and β-arrestin 2 protein expression were examined.Result：Infarct size and left ventricular weight/body weight ratio had no significant difference between MI and carvedilol groups.Compared with sham group,LVESP,positive and negative LV dP/dt were reduced（P0.05）,and non-infarcted myocardial β-arrestin 2 protein level and non-infarcted myocardial collagen volume fraction were increased（P0.05） in MI group.Compared with MI group,carvedilol treatment increased LVESP,reduced LVEDP（P0.05） and improved positive and negative LV dP/dt after MI（P0.05）.Carvedilol treatment also increased non-infarcted myocardial β-arrestin 2 protein levels（P0.05）.Non-infarcted myocardial collagen volume fraction was no differences between MI and carvedilol groups.Conclusion：Carvedilol can improve hemodynamics after MI in rats,probably due to the effect of increase non-infarcted myocardium β-arrestin 2 protein expression.
