摘 要:目的探讨超声辐照载诺帝脂质微泡对正常血管内皮细胞增殖的影响及体外内皮细胞三维成型的抑制作用研究。方法体外培养人脐静脉内皮细胞(HUVEC),并且进行内皮细胞体外三维培养。将细胞分5组,即对照组,载诺帝微泡组,诺帝溶液组,超声辐照诺帝溶液组,超声辐照载诺帝脂质微泡组。四甲基偶氮唑蓝(MTT)法观察不同处理组对细胞生长的影响。比较不同处理组对内皮细胞三维培养的影响。结果在诺帝浓度为200μmol/L时,超声辐照载诺帝微泡组对细胞增殖抑制作用强于其他处理组及对照组;在三维培养3、5、7、9 d后观察发现,超声辐照载诺帝微泡组对体外管腔形成的抑制作用更显著,差异有统计学意义。结论超声辐照载诺帝脂质微泡造影剂对人脐静脉内皮细胞的生长和体外管腔形成有明显抑制作用。Objective To study the effect of inhibition on HUVEC proliferation and the the tubular structure formation by of ultrasound mediated Nordy-carrying microbubbles. Methods HUVEC were cultured in vitro, and divided into 5 groups: control group, simple Nordy microbubbles group, simple Nordy group, Nordy combined with ultra- sound group and Nordy microbubbles combined with ultrasound group. Cell proliferation was measured by MTT as- say. The three-dimensional culture of vascular endothelial cells in vitro was used to observe the effect of lipid microbub- hies containing Nordy on the formation of tubular structures. Results Inhibition of cell proliferation was more obvious- ly at 200 vmol/L in nordy microbubbles combined with ultrasound group. Differences of inhibition of the three-dimen- sional culture observed at 3 d,5 d,7 d,9 d,with other groups were significant. Conclusions Nordy-carrying microbub- bles combined with ultrasound may inhibit the proliferation of HUVEC and the formation of tubular structure in vitro.
