摘 要:目的对四角蛤蜊抗氧化活性进行评价研究。方法采用水提醇沉的工艺路线分别得到四角蛤蜊总提液(MV)、醇沉沉淀(MP)及醇沉上清液(MS)。采用背部皮下注射D-半乳糖建立小鼠衰老模型,评价MV、MP及MS对模型小鼠肝脏与血清中SOD、MDA、GSH-PX、CAT各项指标的影响。以不同极性溶剂对MS萃取,得到石油醚、乙酸乙酯、正丁醇与水部位,评价各部位清除DPPH自由基、清除超氧自由基、螯合和还原铁的能力。结果给予MV、MP、MS的小鼠肝组织SOD、GSH-PX、CAT显著升高(P<0.05),MDA显著降低(P<0.05),血清的SOD、GSH-PX显著升高(P<0.05),MDA显著降低(P<0.05)。MS萃取水部位对DPPH自由基清除、超氧自由基清除和还原能力最强;MS萃取乙酸乙酯部位螯合能力最强。结论四角蛤蜊具有较好的抗氧化活性,其活性较强部位是MS萃取水部位。Objective To evaluate the antioxidative activity of Mactra veneriformis.Methods By adopting the process line of water extraction and alcohol precipitation,we got the total extract(MV),alcohol precipitation(MP)and alcohol supernatant(MS).Except the control group,all of the other groups were subjected to subcutaneous injection of D-galactose solution at the back.The mice models of aging were treated with MV,MP and MS for the observation of their effects on hepatic and serum activities of superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSH-PX),and catalase(CAT).MS was extracted with different polar solvents,and then we got the extracts of petroleum ether,acetic ether,n-butanol and water.The activities of MV extracts on scavenging DPPH free radicals and superoxide radicals,and on iron chelating and reducing were verified.Results After treatment with MV,MP and MS,the activities of hepatic SOD,GSH-PX and CAT exhibited a significant increase(P 0.05) while the activity of MDA was decreased in the liver(P 0.01),and the activities of GSH-PX and SOD in the serum exhibited an obvious increase(P 0.05)but serum MDA activity was decreased(P 0.05).Furthermore,water extract of MS exhibited the strongest scavenging efficacy on O2-and DPPH as well as iron reduction,,and acetic ether extract showed the optimal chelating ability.Conclusion Mactra veneriformis performs certain antioxidant activity in vivo and in vitro,and its effective parts exist in water extract part of MS.
