摘  要:目的研究中药复方回乳抑增一号的HPLC指纹图谱,并分析方中各单味药对全方指纹图谱的贡献度,为复方物质基础和配伍规律研究奠定基础。方法采用高效液相色谱法制备复方及单味药的指纹图谱,色谱条件为:Wondasil C1(84.6mm×250mm,5μm)柱,流动相为乙腈-0.78%NaH2PO4缓冲液(含2%乙酸),梯度洗脱,流速为1 mL/min,检测波长为290 nm,柱温为25℃。结果建立了同一色谱条件下回乳抑增一号复方及单味药的指纹图谱,复方主要特征峰均找到来源的单味药,且复方中丹参酮ⅡA等成分与对应单味药色谱峰在峰高、峰面积上有所差异。10批回乳抑增一号复方样品的相似度均在0.9364以上。结论建立的HPLC指纹图谱相似度高。复方混煎比单煎在化学成分上有质或量的变化。Objective To study the HPLC fingerprint of No.1 Huiru Yizeng and to analyze the effect of single herb from the compound on the fingerprint of whole prescription,thus to supply evidence for further study of material basis and compatibility regularity of the compound.Methods HPLC was used to establish fingerprints of the compound and single herb.The separation was performed on Wondasil C18(4.6mm×250mm,5μm) column with mobile phase consisting of acetonitrile-0.78% NaH2PO4 buffer solution(including 2% glacial acid,by gradient elution) at flow rate of 1.0mL·min-1.Detection wavelength was set at 290 nm and column temperature was 25℃.Results The fingerprints of the compound and single herb have been established under the same chromatographic conditions.The main characteristic peaks were all found in the single herb.The components such as Tanshinone ⅡA in the compound formula was different at peak height and peak area when compared with the corresponding single herb.The similarity of fingerprints of 10 samples was over 0.9364.Conclusion The established HPLC fingerprints have high similarity.The quality or quantity of chemical compositions in the mixed decoction of the compound formula differs from that of the decoction of single herb,which indicates that traditional Chinese medicinal compound is more reasonable.
