摘 要:【目的】观察陈氏内异丸对子宫内膜异位症(endometriosis,EMs)大鼠异位内膜基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达的影响,探讨陈氏内异丸治疗EMs的作用机制。【方法】选用SD大鼠,随机分为假手术组,模型组,陈氏内异丸高、中、低剂量组(中药高、中、低剂量组,剂量分别为7.2、3.6、1.8 g.kg-1.d-1),丹那唑组(剂量为36 mg.kg-1.d-1)。复制EMs大鼠模型,造模成功后各组分别给予陈氏内异丸高、中、低剂量药液及丹那唑混悬液、蒸馏水等灌胃治疗4周。末次给药1周后剥取大鼠内异病灶组织检测MMP-2、MMP-9在异位内膜的表达。【结果】模型组大鼠MMP-2及MMP-9表达强度显著高于中药各剂量组和丹那唑组,差异均有统计学意义(P<0.01);中药高剂量组MMP-2及MMP-9表达强度显著低于中药中、低剂量组及丹那唑组,差异有统计学意义(P<0.05或P<0.01);中药中、低剂量组与丹那唑组比较差异均无统计学意义(P>0.05)。【结论】陈氏内异丸治疗EMs的作用与其能有效抑制大鼠异位内膜组织异常升高的MMP-2和MMP-9的表达有关。Objective To evaluate the effect of Chen's Neiyi Pill(CNP) on expression of the matrix metalloproteinases 2 and 9(MMP-2 and MMP-9) in ectopic endometrium of endometriosis rats,and to explore its possible therapeutic mechanism.Methods SD rats were randomized into pseudo operation group,model group,danazol group(36 mg·kg-1·d-1),and high-,middle-and low-dose CNP groups(in the dose of 7.2,3.6,and 1.8 g·kg-1·d-1 respectively).Rat models of endometriosis were established.After modeling,the medication groups were given the corresponding drugs according to the experimental design,and the pseudo operation group and the model group were given distilled water for 4 weeks.One week after the last medication,ectopic endometrium of endometriosis rats was taken out for the observation of MMP-2 and MMP-9 expression.Results MMP-2 and MMP-9 expression levels were higher in the model group than those in CNP groups and danazol group,the difference being significant(P〈0.01).MMP-2 and MMP-9 expression levels were lower in high-dose CNP group than those in middle-and low-dose CNP groups and danazol group,the difference being significant(P〈0.05 or P〉0.01).The differences of MMP-2 and MMP-9 expression levels were insignificant between middle-and low-dose CNP groups and danazol group(P0.05).Conclusion The therapeutic mechanism of CNP for endometriosis is probably related with the inhibition of the increase of MMP-2 and MMP-9 expression levels in ectopic endometrium of endometriosis rats.
