首乌益智胶囊对血管性认知障碍大鼠脑内血管内皮生长因子(VEGF)影响的实验研究

摘　　要：目的观察首乌益智胶囊对血管性认知障碍(VCI)模型大鼠血管新生的影响。方法用改良的大脑中动脉栓塞法制造血管性认知障碍模型,随机分为模型组、脑复康组及首乌益智胶囊组,每组20只,并设假手术组20只。分别连续给药4周后,应用免疫组织化学法测定缺血灶边缘微血管数和应用western blot法分析大鼠脑内VEGF的表达。结果 (1)血管计数:首乌益智组>脑复康组>模型组>假手术组;各组与假手术组相比,差异均有统计学意义(P<0.01);与模型组比较,脑复康组、首乌益智组差异均有统计学意义(P<0.01);与脑复康组比较,首乌益智组差异无统计学意义(P>0.05)。(2)蛋白表达:首乌益智组>脑复康组>模型组>假手术组;各组与假手术组相比,差异均有统计学意义(P<0.01);与模型组比较,脑复康组、首乌益智组差异均有统计学意义(P<0.01);与脑复康组比较,首乌益智组差异有统计学意义(P<0.05)。结论首乌益智胶囊能明显增强VCI模型大鼠缺血灶边缘新生血管密度,促进脑内VEGF的表达,从而诱导血管再生,以此发挥对脑缺血后的修复和保护作用。Objective To observe the effect of shouwu yizhi capsules on angiogenesis for the rats with vascular cognitive impairment（VCI）. Methods The modified method of middle cerebral artery occlusion （MCAO）was adopted to prepare the model of VCI. By randomization, a model group, a naofukang group and a shouwu yizhi capsules group were divided ,20 rats in each one. Additionally, a sham -operation group （20 rats） was set up. After continuous medication for 4 weeks,immunohistochemical assay was applied to determine the number of micro - vessels on the margin of the foci. Western blot method was adopted to analyze the expression of vascular endothelial growth factor （VEGF） in the brain of rat. Results （ 1 ） Vessel count ： shouwu yizhi group 〉 naofukang group 〉 model group 〉 sham - operation group. As compared with sham - operation group, the results in the other groups presented significant difference （ P 〈 0.01 ）. As compared with model group, the results in naofukang group and shouwu yizhi group presented significant difference （P 〈 0.01 ）. As compared with naofukang group, the result in shouwu yizhi group did not present statistical significant difference （ P 〉 0.05 ）. （2） Protein expression ： shouwu yizhi group 〉 naofukang group 〉 model group 〉 sham - operation group. As compared with sham - operation group, the results in the other groups presen- ted significant difference（P 〈 0.01 ）. As compared with model group, the results in naofukang group and shouwu yizhi group presented significant difference （P 〈 0.01 ）, As compared with naofukang group, the result in shouwu yizhi group presented statistical significant difference （ P 〈 0.05 ）. Conclusion Shouwu yizhi capsules enhance obviously the density of fresh vessels on the margin of ischemic loci in VCI model rats, promote the expression of VEGF in the brain and induce angiogenesis. As a result, the effect of the medicine on the repair and protection after brain ischemia is brought into play.
