针刺对不同时段睡眠剥夺大鼠模型行为学及TNF-α含量的影响

摘　　要：目的探讨失眠的发病机制及针刺治疗失眠的调控机制。方法将SD大鼠随机分为10组,每组8只,设为正常组,失眠组,西药组,针刺组,后三组又根据24,72和120 h三个不同时段各分为三组。各实验组大鼠在睡眠剥夺结束后,根据不同组别和时段(24、72、120 h)分别进行旷场实验,测试其水平得分、垂直得分和中央格停留时间;随即将相应组别大鼠快速摘眼取血,采用放射免疫法检测血浆肿瘤坏死因子(TNF-α)含量的变化。结果①行为学变化:与各时段失眠组大鼠比较,针刺组大鼠水平得分、垂直得分呈下降趋势,针刺组大鼠中央格停留时间呈上升趋势,其中针刺120 h组垂直得分和中央格停留时间有统计学意义(P<0.01)。②血浆TNF-α含量变化:与各时段失眠组比较,针刺组血浆TNF-α含量整体呈升高趋势,其中针刺72 h组和120 h组的TNF-α含量明显升高(P<0.05,P<0.01)。结论针刺能显著改善睡眠剥夺大鼠的精神状态,增强其认知能力和反应能力;针刺可以提高睡眠剥夺大鼠TNF-α的含量,达到缓解失眠的效果,且治疗时间越长,效果越明显。Objective To explore the mechanism of insomnia mice for the purpose of providing some experimental proof about acupuncture therapy on insomnia. Methods 80 male SD mice were randomly divided into control （CC） , model （S）, insomnia with western medicine （SX）, insomnia with acupuncture （SZ） groups. Except mice in control group, mice in other groups were divided into 3 groups according to the different time of sleep deprived： 24, 72 and 120 hours. Content of TNF-α in the eye of mice was detected by radiation immune method. The clinical effect and the influence on open field test （testing：crossing score, rearing score, latency time） were studied. Results In the behavior test, the crossing score and rearing score of SZ group were significantly less than that of CC group, and the latency time was raised. In the test of immune cytokine, TNF-α of acupuncture group for 72 h and 120 h （SZ group） was significantly more than those of CC group （P 〈 0. 01, P 〈 0. 05 ）. Conclusions Acupuncture has improved behavior effects as reaction and learning ability, and can increase the TNF-α content and decrease the damage of insomnia.
