摘  要:目的观察补阳还五汤对糖尿病大鼠血脂及主动脉中过氧化物酶体增殖物激活受体(PPAR)-γ表达的影响。方法 Wistar雄性大鼠随机分为正常对照组和造模组。造模组大鼠尾静脉注射链脲佐菌素(STZ)溶液(30mg/kg);将造模成功的糖尿病大鼠随机分为糖尿病模型组(DM组)、二甲双胍组(MF组)、补阳还五汤高剂量组(BH组)、补阳还五汤低剂量组(BL组),分别给予灌服等容积蒸馏水、二甲双胍混悬液0.75g/(kg.d)、高剂量补阳还五汤22.5g/(kg.d)、低剂量补阳还五汤5.4g/(kg.d)。正常对照组给予注射等量枸橼酸缓冲液。连续给药6周后,检测糖尿病大鼠灌胃后血糖(BG)、胆固醇(TC)、甘油三酯(TG)及免疫荧光染色测定主动脉PPAR-γ表达。结果 BH组能降低BG、TC、TG的水平,并能明显增加糖尿病大鼠主动脉PPAR-γ表达。结论补阳还五汤能调节糖尿病大鼠糖脂代谢,增加糖尿病大鼠血管PPAR-γ表达。Objective:To investigate the effects of Buyanghuanwu Decoction on blood glucose,blood fat and PPAR-γ expression in diabetic rats.Methods: Male Wistar rats were randomly divided into the normal group and the model group.The model group was given intravenous injection of streptozotocin(STZ) solution(30mg/kg) to make DM rats.The DM rats were randomly divided into DM model group(DM group),The metformin group(MF group),the Buyanghuanwu Decoction high-does group(BH group) and the Buyanghuanwu Decoction low-does group(BL group).These groups were respectively given the distilled water,the metformin with 0.75g/kg.d and the Buyanghuanwu Decoction with 22.5g/kg.d or 5.4g/kg.d.The normal group was given equal citrate buffer.The treatment was lasted for 6 weeks to measure the changes in blood glucose,blood fat and the expression of PPARγ.Results: Buyanghuanwu Decoction significantly reduced BG,TC,TG levels and increased the expression of PPAR-γ in the aortic sinus.Conclusion: Buyanghuanwu Decoction can prevent diabetes vascular complications by regulation of glycolipid metabolism and increase PPAR-γ expression.
