摘 要:目的:研究既有建筑环境甲醛吸入对小鼠肺组织及炎性细胞因子表达的影响。方法:采用胶合板、粘合剂自制染毒室,染毒室甲醛平均浓度分别为0.28、0.73和1.21mg/m3。染毒组小鼠每日染毒1次,每次4h,连续8d。8d后,取肺组织,HE染色,观察肺组织病理学改变,免疫组织化学方法观察肺组织TNF-α、IL-17、IL-6的阳性表达。结果:甲醛吸入组小鼠出现肺泡壁毛细血管扩张、充血,炎细胞浸润的病理改变;甲醛染毒组(0.28、0.73和1.21mg/m3)小鼠肺组织TNF-α、IL-17表达明显高于正常对照组(P<0.05);甲醛染毒组(0.73和1.21mg/m3)小鼠肺组织IL-6表达较正常对照组明显增高(P<0.01)。结论:既有建筑环境甲醛可致小鼠肺组织病理损伤,炎性细胞因子TNF-α、IL-6、IL-17表达明显增强,提示炎性细胞因子参与了甲醛致肺组织损伤的病理发展过程。To investigate the effects of the existing building environment of formaldehyde inhalation on the histopathology of lung in mice and the expression of inflammatory cytokines,the enclosure chambers were made by plywood and adhesives,and the mice in formaldehyde treatment groups were exposed to 0.28,0.73,1.21mg/m3 concentration by inhalation in the chambers for 4h per day and continuously for 8 days.Then,the lung tissue was obtained and the pathologic identification was done by HE staining and the expression of TNF-α,IL-17,IL-6 in the lung tissue was detected by immunohistochemistry method.In comparision with the control group,formaldehyde inhalation groups showed that the pathological changes were angiotelectasis,congestion in alveolar wall and inflammatory cell infiltration;the expression of TNF-α and IL-17 in the lung tissue in formaldehyde exposed groups(0.28,0.73,1.21mg/m3)was obviously higher than that in the control group(P0.05);in comparison with control group,the expression of IL-6 in the lung tissue in formaldehyde exposed groups(0.73,1.21mg/m3)showed a significant increasing(P0.01).It is concluded that existing building environment of formaldehyde inhalation can induce the pathological damages in the lung of mice;the level of inflammatory cytokines(TNF-α,IL-6,IL-17) in the lung tissue are significantly increased,so as to point out that the inflammatory cytokines(TNF-α,IL-6,IL-17) participate in the process of the pathological damages in the lung tissue induced by formaldehyde.
