摘　　要：目的:研究男性年龄对人精子氧化应激水平、核DNA损伤的影响。方法:选择因女方原因行体外受精-胚胎移植的男性精液标本95例,按男方年龄分为3组,年龄<35岁组40例、35～39岁组34例,≥40岁组21例,观察不同年龄对人精子氧化应激水平、核DNA损伤情况。结果:3组精液量、精子浓度、精子正常形态差异均无统计学意义(P>0.05),≥40岁组的a+b级精子的百分率低于<35岁组(P<0.05),精浆活性氧水平为(17.66±7.73)%,高于<35岁组和35～39岁组(均P<0.05),精子DNA损伤率为(21.12±11.01)%高于<35岁组(P<0.05)。经Pearson相关系数分析,精子氧化应激水平、核DNA损伤与男性年龄均呈正相关(r分别为0.43和0.27,P<0.05)。结论:男性年龄增加可能导致前向运动精子比率降低,精子氧化应激水平、核DNA损伤升高。精子氧化应激水平、核DNA损伤与男性年龄存在正相关。在生育过程中,男性年龄因素应引起临床关注。Objective ： To explore effects of age on oxidative stress and DNA integrity of human sperm. Methods ： Ninety - five men aged 21 to 53 who underwent IVF - ET for female infertility were divided into three groups according to their ages. Semen samples were collected. The sperm parameters, maleic dialdehyde （MDA） concentration and nuclear DNA integrity of the three groups were analyzed . Results： There were no significant differences in the semen volume, sperm concentration and sperm normal morphology among the three groups （P 〉 0.05 ）. Men who were less than 35 years old and 35 - 39 years old had significantly more motile sperm than those not less than 40 years of age （P 〈 0.05）. The oxidative percent of semen samples of men not less than 40 years old was higher than those in men less than 35 and 35 - 39 years old （ P all 〈 0.05 ）. The DNA damaged percent of semen samples of men not less than 40 years old was higher than those in men less than 35 （P 〈 0.05） and 35 - 39 years old （ P 〉 0.05 ）. The reactive oxygen species level and DNA integrity were positively correlated with the age by Pearson correlation analysis （ r = 0. 43, and 0.27, respectively, P 〈 0.05 ）. Conclusion ： Male age probably affects motile, oxidative stress and sperm DNA damage. It is necessary to concern about the male age when preparing for pregnancy.
