摘 要:目的观察补肾活血中药对大鼠慢性高眼压(elevated intraocular pressure,EIOP)模型外侧膝状体损伤的干预作用,探讨其作用机理。方法采用烙闭上巩膜静脉法建立大鼠慢性EIOP模型,随机分为3组:空白组、模型组、给药组,观察补肾活血中药对EIOP大鼠眼压、外侧膝状体(lateral geniculate nucleus,LGN)病理形态学变化的影响。结果本实验采用的烙闭上巩膜静脉的造模方法使大鼠眼压明显升高,与造模前比较差异有显著统计学意义(P<0.01),在造模后8周(实验结束)眼压仍为造模前的2.5~3.0倍;LGN病理切片半定量分析表明:模型组神经元细胞密度(51.14±10.52)μm-2、神经元剖面积密度(6.57±1.59)μm-2、有髓纤维密度(0.3556±0.1019)μm-2、Nissl小体积分光密度31826±3796,与空白组比较差异均有统计学意义(P<0.01或0.05)。模型组Nissl小体积分光密度低于给药组(33906±3213),给药组神经元细胞密度(54.27±12.70)μm-2与空白组(60.95±13.35)μm-2比较差异有显著统计学意义(P<0.01),给药组有髓纤维密度(0.5611±0.1385)μm-2与模型组比较,差异有统计学意义(P<0.05)。结论补肾活血中药有助于EIOP大鼠LGN损伤的修复。Objective To observe the effect of traditional Chinese medicine(TCM) of BuShenHuoXue on the pathological changes of lateral geniculate nucleus(LGN) injury in rat model of chronic elevated intraocular pressure(EIOP),and preliminarily explore its mechanism.Methods The rat model of chronic EIOP was established by unilaterally cauterizing three episcleral vessels,then randomly divided into 3 groups:control group,model group and treatment group.The effect of TCM drugs of BuShenHuoXue on the EIOP rat model's intraocular pressure(IOP) and pathological changes of LGN were observed.Results Unilaterally cauterizing episcleral vessels increased IOP of the rat model obviously,there was significant difference compared with pre-operation(P0.01).At the end of the experiment(8 weeks),the level of IOP was 2.5-3.0 times than before modeling;Semi-quantitative pathological analysis showed that the density of neuron cells in the model group was(51.14±10.52)μm-2,cell density of sectional area was(6.57±1.59)μm-2,density of myelinated fiber was(0.355 6±0.101 9)μm-2,integrated optical density of Nissl body was 31 826±3 796,compared with the control group,there were statistical differences(P0.01 or 0.05).There was statistical difference in integrated optical density of Nissl body between the model group and treatment group 33 906±3 213(P0.05),significant difference in the density of neuron cells between the treatment group(54.27±12.67)μm-2 and control group(60.95±13.35)μm-2(P0.01),statistical difference in density of myelinated fiber between the treatment group(0.561 1±0.138 5)μm-2 and model group(P0.05).Conclusion TCM of BuShenHuoXue can repair the LGN injury in the rat model of chronic EIOP.
