摘 要:[目的]探讨生长相关蛋白(Gap-43)在电针治疗大鼠脊髓损伤(SCI)中的变化及意义.[方法]以2个月龄Wistar大鼠(96只)为受试对象,随机分为对照组(A组)、损伤组(B组)、治疗对照组(C组)和电流刺激治疗组(D组).B、C、D组于T9处行脊髓全横断,D组予电针治疗.用Western-blot测定各组损伤段脊髓中Gap-43不同时相点表达量及其变化,以苏木精-伊红和免疫组化染色对受损脊髓进行形态学观察.[结果]SCI后Gap-43有少量表达,经过电针治疗后,表达显著增多.[结论]Gap-43是SCI后神经纤维再生的重要因子,电针治疗对其表达具有明显的促进作用.[Objective] To explore the change of growth associated protein(Gal〉43) in rats of spinal cord injury (SCI) treated with electro-acupuncture and its significance. [Methods] Totally 96 Wistar rats with 2-month age were selected as the subjects and randomly divided into the control group(group A), SCI group(group B), the sham-treatment group(group C) and the electro-acupuncture treatment group(group D). Group B, C and D underwent total-transect SCI at the vertebrae level of T9. Group D was treated with electro-acupuncture. Western-blot was used to examine the expression and change of Gap-3 in injured spinal cord at different time points. Hematoxylin-feosin and immunohistochemical staining methods were used to observe the morphology of injured spinal cord. [Results]A little expression of Gap-43 after SCI was found. After electro-acupuncture treatment, the expression of Gap-43 increased obviously. [Conclusion] Gap-43 is an important factor of nerve fiber regeneration after SCI. Electro-acupuncture can obviously promote the expression of Gap- 43 protein.
