赤丹丸中芍药苷在大鼠体内各组织的分布

摘　　要：目的建立芍药苷在大鼠组织中的高效液相色谱法(HPLC)检测。比较配伍前后大鼠各组织中芍药苷浓度的变化,考察赤丹丸中芍药苷在大鼠各组织分布状况的改变。方法建立HPLC法测定芍药苷在大鼠组织中的含量。色谱仪:Agilent 1100;色谱柱:Diamonsil C18(150mm×4.6mm,5μm);检测波长:230nm;流速:1mL/min;流动相:甲醇-磷酸水梯度洗脱。结果赤芍组中芍药苷在各组织中的分布特征为肺>肾>卵巢>子宫>脾>心>肝,赤丹丸组中芍药苷在各组织中的分布特征为肺>子宫>肾>心>卵巢>肝>脾。赤丹丸组大部分组织中芍药苷含量大于赤芍组,子宫和卵巢更为明显。结论芍药苷在各组织中的浓度较赤芍组明显升高,赤丹丸的配伍作用在一定程度上促进芍药苷在组织中的分布。子宫和卵巢中芍药苷的含量也较大。Objective To establish the HPLC method for detecting paeoniflorin in rat plasma and tissues. To study the pharmacokinetic changes of paeoniflorin in Chidan pills by testing the concentration of paeoniflorin in plasma. To investigate the changes of tissue distribution when co-administrated with other ingredients in Chidan pills. Methods HPLC method for analysis of paeoniflorin in plasma and tissues is established. HPLC analyses were conducted by using an Agilent 1100 and Diamonsil C18 （150 mm× 4.6 mm, 5 μm） column. The flow-rates of mobile phases were maintained at 1 mL/min, and the detection was performed at the wave length of 230 nm. The mobile phases were methanol and water with phosphoric acid 0. 1% by gradient solution. Results The tissue dis- tribution characteristics of paeoniflorin in rat tissues is from lung〉kidney〉ovary〉uterus〉spleen〉heart〉liver to lung〉uterus〉kidney〉heart〉ovary〉liver〉spleen after co-administration of other ingredients in Chidan pills. Most tissues＂ concentrations of paeoniflorin in Chidan pills group are higher than those in Radix Paeoniae Rubra group, especially in uterus and ovary. Conclusion The tissue concentration of paeoniflorin is markedly enhanced after co-administration of other ingredients in Chidan pills. The concentrations of uterus and ovary are considera- ble. As a result, we can provide the thoughts for the mechanism of the Chinese traditional medicine compound preparation in vivo and guide the clinical use of medicines.
