摘 要:目的探讨具有雌激素活性的牛膝的有效中药单体蜕皮甾酮(ecdysterone,ECR)对过氧化氢(H2O2)诱导氧化损伤的人晶状体上皮细胞(human lens epithelial cell,HLEC)核因子κB(nuclear factor-kap-paB,NF-κB)p65表达的影响。方法实验分5组:正常组、H2O2组、雌二醇(E2)组、ECR组、核因子抑制剂二硫代氨基甲酸吡咯烷(pyrrolidine dithiocarbamate,PDTC)组;采用流式细胞术(FCM)测定各组标本中HLEC的NF-κBp65的表达率。结果正常HLEC能表达一定的NF-κBp65(9.53%),H2O2组NF-κBp65的表达率明显增高(39.87%,P<0.01),PDTC组则明显降低(5.90%,P<0.01),ECR组(13.99%)和E2组(25.18%)介于正常组与H2O2组之间,均低于H2O2组(P<0.01)。结论 H2O2可以诱导HLEC的NF-κB的活化表达,具有雌激素活性的中药单体ECR可以有效抑制NF-κB的活化表达。Objective To study the effects of ecdysterone (ECR) on the expression of nuclear factor (NF)-KB in H2O2induced oxidative damage of human lens epithelial cells (HLECs). Methods The cultured HLECs were divided into 5 groups, i.e., the control group, the H2O2 group, the β-estradiol (E2) group, the ECR group, and the pyrrolidine dithiocarbamate group (PDTC) group. The expression rate of NF-KB p65 in the HLECs were detected by flow cytometer (FCM). Results The expression of NF-KB p65 occurred in normal HLECs (9. 53%). The expression rate of NF-KB 1065 in the H202 group obviously increased (39. 87%, P〈0. 01 ). The expression rate of NF-KB p65 in the PDTC group obviously decreased (5. 90%, P〈0. 01 ). The expression rates of NF-KB p65 in the ECR group (13.99%) and the E2 group (25.18%) ranged between the control group and the H202 group, but still lower than that of the H202 group (P〈0.01). Conclusions The activation of NF-KB in the HLECs could be induced by H2O2. ECR with the estrogenic activity could effectively inhibit the activation of NF-KB.
