摘  要:目的探讨氢氧化锌[Zn(OH)2]对IPV-HBV联合疫苗诱导小鼠体液免疫应答的影响。方法将ICR小鼠随机分为4组:生理盐水对照组、IPV(Ⅰ、Ⅱ、Ⅲ型各1×107pfu)-HBV(2μg)+Zn(OH)(20.5 mg)组、IPV(Ⅰ、Ⅱ、Ⅲ型各1×107pfu)-HBV(2μg)+A(lOH)(30.5 mg)组和IPV-HBV组,经大腿肌肉内侧免疫2次,间隔4周。分别于初次免疫后第4、8、12、16、20和24周采血,分离血清,ELISA法检测小鼠血清中各抗原的特异性IgG抗体水平。结果各实验组相对于对照组,在初次免疫后4周时均产生针对各抗原的特异性IgG抗体;8周时达最高,并能持续较长时间。IPV(Ⅰ、Ⅱ、Ⅲ型各1×107pfu)-HBV(2μg)+Zn(OH)2组产生的特异性IgG抗体水平均显著高于其他各组(P<0.05或P<0.01)。结论 Zn(OH)2能明显增强IPV-HBV联合疫苗诱导的小鼠体液免疫应答,其免疫佐剂效应优于A(l OH)3佐剂。Objective To investigate the effect of zinc hydroxide [Zn (OH)El on IPV-HBV combined vaccine-induced humoral immune response. Methods ICR mice were randomly divided into one control and three test groups, and immunized i. m. twice with physiological saline, IPV (types I , lI and HI , each 1 x 107 pfu)-HBV(2 p^g) + Zn(OH)2 (0. 5 mg), IPV (types I , II and II1 , each 1 x 107 pfu)-HBV(2 μg) + Zn(OH)2 (0. 5 mg) and IPV-HBV respectively, at an interval of 4 weeks. Serum samples were collected at weeks 4, 8, 12, 16, 20 and 24 after the first immunization, and determined for specific IgG levels against various antigens by ELISA. Results As compared with those in control group, specific IgG against various antigens were induced in three test groups at week 4 after the first immunization, of which the levels reached peak values at week 8 and lasted for a long time. However, the specific IgG level in IPV (types I , Ⅱ and Ⅲ , each 1 x 107 pfu)-HBV (2 μg) + Zn (OH)2 group was significantly higher than those in other groups (P 〈 0. 05 or P 〈 0. 01). Conclusion Zn (OH)2 enhanced IPV-HBV combined vaccine-induced humoral immune response significantly, of which the adjuvanticity was superior to that of AI(OH)z.
