芹菜素对人鼻咽癌CNE-2Z细胞化疗敏感性的增强作用

摘　　要：目的观察芹菜素(AP)能否增强人鼻咽癌CNE-2Z细胞对顺铂(DDP)的化疗敏感性。方法采用MTT法对CNE-2Z细胞的增殖抑制率进行检测,中效原理法对联合用药的相互作用进行分析判断;采用流式细胞术和PI/Ho-echst33258荧光双染法检测细胞凋亡;RT-PCR法检测凋亡相关基因bcl-2、bax mRNA表达的改变。结果 AP、DDP单用及合用均明显抑制CNE-2Z细胞的生长,且呈剂量依赖性关系,两药合用时在中、高浓度呈协同效应;流式细胞术和荧光双染法结果表明低浓度AP联合DDP更能明显促进肿瘤细胞的凋亡;与单独用药组比较,RT-PCR结果显示联合用药组bcl-2 mRNA表达明显降低,而bax mRNA表达明显增高。结论芹菜素可增强顺铂对CNE-2Z细胞增殖的抑制作用和诱导凋亡作用,其机制可能与上调bax和下调bcl-2的表达,导致Bcl-2/Bax比值下降有关。Aim To observe the chemosensitization effects of apigenin on cisplatin in human nasopharyngeal carcinoma CNE-2Z cells.Methods The inhibitory effects of apigenin,cisplatin and apigenin combined with cisplatin on the growth of CNE-2Z cells were determined by MTT assay.The combination effect was determined by using the method of median-effect principle.Cell apoptosis was detected by flow cytometry and Hoechst3358/PI fluorescent staining.The changes of bcl-2,bax mRNA level were quantified by RT-PCR.Results Apigenin and cisplatin（individual or combined） significantly inhibited the growth of CNE-2Z cells in a dose-dependent manner.The interaction of the two drugs was synergistic at medium or high concentrations.The results of flow cytometry and PI/Hoechst33258 fluorescent staining showed that the combinative application group（at low concentrations） could significantly induce cells apoptosis.Compared to apigenin or cisplatin treatment group,the mRNA expression of bax was obviously up-regulated as well as that of bcl-2 was significantly down-regulated in the combination group.Conclusions Apigenin promotes proliferative inhibition and apoptotic induction of CNE-2Z cells by cisplatin.The mechanism may associate with the up-regulation of Bax expression and the down-regulation of Bcl-2 expression,which results in a decline in Bcl-2/Bax ratio.
