摘 要:目的:研究两种基于免疫反应和电化学的简便、灵敏、特异检测内毒素的新方法。方法:应用ELISA建立PMB-ET-ELISA法和免疫传感器法对大肠杆菌内毒素进行分析,并对两种方法检测范围和最低检测限进行分析和比较。结果:ELISA方法检测精制内毒素范围为1μg/ml~1 mg/ml,检测大肠杆菌O157:H7范围为105 CFU/ml~109 CFU/ml,相当于内毒素190μg/ml~2918μg/ml。免疫生物传感器法检测精制内毒素范围为1μg/ml~1 mg/ml;检测大肠杆菌O157:H7范围为106 CFU/ml~109 CFU/ml,相当于内毒素288μg/ml~1416μg/ml。结论:建立的ELISA和免疫传感器两种检测内毒素新方法有效、稳定、可用。Objective:To study two electrochemical methods for the detection of endotoxin based on simpleness,sensitivity,and specificity.Methods: Escherichia coli endotoxin were analyzed by PMB-ET-ELISA and immunosensor.Moreover,the detection range and the detection limit of the two methods were analyzed and compared.Results: With ELLSA(enzyme-linked immunosorbent assay),the detection range of refined endotoxin were 1 μg/ml^1 mg/ml,E.coli O157: H7 detection range were 105 CFU/ml^109 CFU/ml,equivalent to endotoxin 190 μg/ml^2918 μg/ml.While with immunosensors,the detection range of refined endotoxin were 1 μg/ml^1 mg/ml;detection range of E coli O157:H7 were 106 CFU/ml^109 CFU/ml,equivalent to endotoxin 288 μg/ml^1416 μg/ml.Conclusion: The ELISA and immunsensor in endotoxin detection are effective,stable and feasible.
