摘 要:以外植体母株为对照,利用ISSR标记对来自同一忽地笑鳞茎诱导,通过不定芽和体胚发生途径形成的再生一代和二代植株的体细胞无性系变异进行了分析。结果表明,再生植株出现不同程度的ISSR位点变异,表现为新增加带和缺失带。体胚发生途径出现的ISSR位点变异率高于不定芽形成途径。第二世代的变异率高于第一世代。体胚发生途径形成的第一和第二世代植株ISSR位点平均变异率分别为0.7%和1.4%,而不定芽途径形成的第一和第二世代植株ISSR位点平均变异率分别为0.4%和0.8%。来自相同途径的同一世代的各单株间,ISSR位点变异率差别较大,在AB1-3、AB1-8和SE1-2中未检测到ISSR位点变异,而AB1-2和SE1-1的ISSR位点变异率分别达到0.6%和1.0%。第一世代发生变异的植株,其再生第二世代植株中这种变异依然存在。The somaclonal variations of the first and second regeneration plants derived from bulb of Lycoris aurea by adventitious bud formation and somatic embryogenesis were analyzed with ISSR markers.The results indicated that there were absence of DNA fragments and newly amplified bands in amplified products of somaclonals.The regeneration plants by somatic embryogenesis showed higher variation frequency than that by adventitious bud formation,and the second regeneration plants had hgher variation frequency than the first regeneration plants.The variation frequency of the first and the second regeneration plants by somatic embryogenesis were 0.7% and 1.4%,while the variation frequency of the first and the second regeneration plants by adventitious bud formation were 0.4% and 0.8%,respectively.There were differences of variation frequency among individuals from one generation by the same way.The variation frequency of AB1-2 and SE1-1 was 0.6% and 1.0%,respectively,but variation was not detected in AB1-3,AB1-8 and SE1-2.Same variation was detected in the second regeneration plants as the first regeneration parent plants.
