摘 要:目的观察人工关节磨损钛微粒的大小及不同微粒一细胞比对成骨细胞低密度脂蛋白受体(Lrr,5)mRNA表达的影响。方法(0.35±0.09)μm磨损钛微粒分别以微粒-细胞比1:1、10:1、100:1、500:1,(1.5±0.38)p.m及(6.254±2.13)μm大小磨损钛微粒以微粒.细胞比10:1与成骨细胞联合培养,应用逆转录.聚合酶链式反应(RT.PCR)技术检测成骨细胞Lrp5mRNA表达。结果随着钛微粒-细胞比增高,成骨细胞Lrp5mRNA表达逐渐减低,当钛微粒-细胞500:1时未检测到kp5mRNA表达,各组间差异有统计学意义(P〈0.05)。在微粒-细胞比10:1时,(0.354-0.09)μm钛微粒组较(1.5±O.38)Ixm和(6.254±2.13)斗m钛微粒组,(1.54±0.38)μm钛微粒组较(6.25±2.13)um钛微粒组,成骨细胞L叩5mRNA表达降低,差异有统计学意义(P〈0.05)。结论人工关节磨损钛微粒可以通过Lrp5信号途径影响骨形成,微粒大小及数量是决定磨损微粒细胞学反应的重要参量,微粒越小,微粒.细胞比越高,对成骨细胞Lrp5mRNA表达的抑制性越强。Objective To approach the effects of size-separated titanium particles and the ratio of titanium particles to osteoblasts on low-density lipoprotein receptor 5 ( Lrp5 ) mRNA in osteoblasts. Methods (0. 35 ±0. 09) Ixm titanium particles were co-cultured with osteoblasts in vitro at various particle number to osteoblasts ratios (1: l, 10: 1, 100: 1,500: 1) and (1.5±0. 38)μm, (6. 25±2. 13) txm titanium particles at rate 10 : 1 with osteoblasts. The co-culture supernatants were analyzed at the mRNA level by using reverse transcriptase polymerase chain reaction (RT-PCR). Results Weard titanium particles sig- nificantly downregulated Lrp5 mRNA expression in osteoblasts. Compared to the normal control group, the expression of Lrp5 mRNA was significantly reduced in osteoblasts in the other groups (P 〈 0. 05). Osteo- blasts co-cultured with (0. 35 ± 0. 09) μm titanium particles, compared to those co-cultured with ( 1.5±0. 38 ) Ixm and (6. 25±2. 13 ) μm titanium particles, significantly restrained Lrp5 mRNA expression (P 〈 0. 05 ). Conclusion The size and concentration of titanium particles are important parameters to decide the cytobiologic model of osteoblasts. Smaller size and higher concentration of weared titanium particles can more significantly inhibitied the Lrp5 mRNA expression.
