摘  要:为了研究血清饥饿和接触抑制2种常用的处理方法对供核细胞周期的影响,试验采用血清饥饿和接触抑制2种方法处理经过纯化的供核细胞,并用流式细胞仪检测处理后的供核细胞的细胞周期,通过Modfit软件分析不同处理方法对细胞周期产生的具体影响。结果表明:血清饥饿和同接触抑制2种方法处理过的细胞停留在G0/G1期和S期的细胞百分数差异不显著,但是同对照组细胞相比差异显著(P<0.05);停留在G2/M期的细胞百分数差异显著(P<0.05),但是同对照组细胞相比差异不显著(P>0.05)。To research the effect of two commonly used methods on the cell cycle of donor cells, the experiment used the two methods of contact inhibition and serum starvation to treat the donor cells after purified, detecting the cell cycle by using flow cytometry. Modfit software was used to analyze the effect of different treatments on the cell cycle. The results showed that the percentage of cells between the two treat groups re- mained at the GO/G1 phase was not significant, but it was higher than that of the control group ( P 〈 0.05 ). The percentage of cells between the two treat groups remained at the S phase was not significant, but it was lower than that of the control group ( P 〈 0.05 ). The results demonstrate that contact inhibition and serum starvation have tremendous influence on cell cycle.
