摘 要:目的:运用郭氏系统血液免疫反应分离实验体系,探讨在抗原激活的情况下异体白细胞、红细胞与自体白细胞、红细胞对反应体系中IL-8、IL-10影响的变化规律。方法:以人类大肠埃希菌作为激活原,加入异体白细胞、红细胞与自体白细胞、红细胞对比,37℃孵育1h,取上清液用ELISA测定IL-8、IL-10的分泌量。结果:IL-8的分泌量异体白细胞组(199.75970±62.743079)与自体白细胞组(465.92697±111.354713)相比明显下降(P=0.003<0.05),异体红细胞组(119.05455±31.056229)与自体红细胞组(132.63303±35.072837)相比无明显差异(P=0.456>0.05);IL-10的分泌量异体白细胞组(16.93582±2.346794)%与自体白细胞组相比(46.16061±9.180075)%明显下降(P=0.017<0.05),异体红细胞组(22.28497±4.010929)与自体红细胞组(27.58494±4.740832)相比无明显差异(P=0.274>0.05)。结论:同种异体同血型白细胞对自体血液免疫反应细胞因子的分泌具有负向的调控作用,同种异体同血型红细胞无此作用。Objective To observe the effects of autogenetic or allogenous white blood cells and erythrocytes on cytokines of IL-8 and IL-10 in immunological reactions activated by antigens by means of natural isolated experimental system. Methods Inactivated Escherichia coli was used as activating antigen, autogenetic white blood cells or allogenous white blood cells were added into the reaction system, and were compared with one another. After incubation an hour in 37℃, supernate fluid is suspended and detected the changes of IL-8, IL-10 by ELISA assay. Results The secretion of IL-8 of allogenous white blood cells experimental group [(199.75970±62.743079)%] is much lower than that of autogeneic white blood cell experimental group [(465.92697±111.354713)%] (P = 0.003 0.05). The secretion of IL-10 of allogenous white blood cells experimental group [(16.93582±2.346794)%] is much lower than that of autogeneic white blood cells experimental group [(46.16061±9.180075)%] (P = 0.017 0.05). The secretion of IL-8 and IL-10 of allogenous erythrocytes and autogenous erythrocytes shows no significant difference. Conclusion Allogenous white blood cells has depressant effects on cytokines in autoblood immune reaction. There's no significant difference of the secretion of IL-8 and IL-10 whether erythrocytes exist or not. This result indicates that allogenous white blood cells transfusion can induce the transfusion related immunoregulation, and allogenous erythrocytes has no this role.
