摘 要:目的:观察葛根异黄酮(TIP)对去卵巢大鼠骨骼质量的影响。方法:选用健康成年SD雌性大鼠40只,随机分为5组:假手术组、模型对照组、低、中、高剂量TIP组;假手术组切除卵巢旁脂肪组织,其余各组均切除双侧卵巢,建立去卵巢大鼠动物模型。按25、50、100 mg/kg 3个剂量的TIP分别给去卵巢大鼠灌胃3个月,实验结束后处死大鼠,用相关方法检测骨骼质量变化的指标。结果:中、高剂量TIP组动物的体重与相对应时相的模型组比较均有下降(P<0.05);中、高剂量TIP组大鼠股骨重量、骨钙含量与模型对照组比较增加(P<0.05);高剂量TIP组大鼠的血清钙含量增高,血清磷、骨钙素和碱性磷酸酶含量均低于模型对照组(P<0.05);中、高剂量TIP组大鼠的股中心BMD和远心BMD均随葛根异黄酮剂量的增加而增加,呈剂量—效应关系,与模型对照组比较差异均有显著性(P<0.05)。结论:一定剂量葛根异黄酮能提高骨钙含量、增加骨密度,对去卵巢大鼠骨骼质量有明显的改善作用。Objective: To investigate Effects of Total isoflavones of pueraria on bone quality in ovariectomized rats. Methods: Choose healthy adult female SD rats were randomly divided into five groups: sham operation group, model control group and Total isoflavones pueraria groups of low, medium,high - dose ; Sham group removal of Ovarian side adipose tissue , the ovariectomized were established rat model of other groups.. Press 25,50,100 mg / kg three doses of Total isoflavones of pueraria, give of ovariectomized rats were fed to three month; the experiment end rats were killed , detected with relevant indicators of bone quality changes. Results: Medium and high doses Total isoflavones group the animal weight and with relative phase model group were decreased ( P 〈 0. 05 ) ; Medium and high do- ses Total isoflavones group femur weight, bone calcium content compared and with the model control group increased (P 〈 0. 05) ; High - dose TIP rats increased serum calcium, serum phosphorus, osteocalcin, and alkaline phosphatase levels were lower than model group (P 〈 0. 05) ; Medium and high doses group of total isoflavones of pueraria rats distal femur BMD and central BMD with both doses of Pu- eraria lobata isoflavones increased in dose - response relationship with the model control group differences were significant (P 〈 0. 05). Conclusion: A certain dose of Pueraria lobata isoflavones can increase bone calcium content, increased bone mineral density in ovariectomized rats significantly improved the quality of the role.
