摘  要:目的通过对损伤的大鼠脊髓早期不同时机减压后测定脊髓组织中热休克蛋白(HSP)70的表达及其与神经细胞凋亡的相关性研究,评价早期减压的疗效。方法采用环扎法建立大鼠脊髓损伤模型,随机将大鼠分为4组,对照组、8h脊髓减压组(实验B组)、72h脊髓减压组(实验C组)和不减压组(实验D组),并分别在1d、3d、7d、14d和21d处死后取各组大鼠受损脊髓进行HE染色,免疫组化法、光密度测量法观察脊髓细胞HSP70的表达,TUNEL法观察神经细胞的凋亡。应用SPSS17.0统计软件对数据进行分析。结果实验组HSP70、TUNEL阳性细胞数及HSP70积分光密度各组组间比较差异均有统计学意义(P<0.05),实验组HSP70表达与TUNEL阳性细胞数的相关性分析表明两者呈正相关(r=0.675~0.921)。结论大鼠脊髓损伤后早期减压,神经细胞凋亡减少,可减轻脊髓损伤;HSP70表达与脊髓损伤神经细胞凋亡的诱导有关,与减压时机无关。Objective To investigate relationship between HSP70 and neuronal apoptosis measured by optical density in rats treated with cerclage spinal cord injury and decompression at different early time,and to evaluate the efficacy of early decompression.Methods Cerclage rat model of spinal cord injury, rats were randomly divided into four groups,divided into control group,eight hours of spinal cord decompression group,72 hours spinal decompression group and non-decompression group,in 1 d,3 d,7 d,14 d and 21 d of each group were killed after spinal cord damage in rats with HE staining, immunohistochemistry,optical density measurement of spinal cord cells, the expression of HSP70,TUNEL apoptosis of nerve cells was observed. SPSS 17.0 statistical software for data analysis.Results Experimental group HSP70,TUNEL-positive cells and HSP70 integrated optical density were significantly different, each group P<0.05 significant difference in HSP70 expression in the experimental group and the number of TUNEL-positive cells compared with the statistics, the two were positively correlated (r=0.675-0.921).Conclusion Early decompression of spinal cord injury can reduce neuronal apoptosis and reduce spinal cord injury;HSP70 expression is related with apoptosis cell induction after spinal cord injury,and no-related with the decompression time.
