摘  要:目的 探讨高压氧(HBO)联合亚低温(MHT)治疗对创伤性脑损伤(TBI)细胞凋亡及Bcl-2表达的影响.方法 制作大鼠自由落体脑挫伤动物模型,设立TBI常规治疗组(TBI组)、常规治疗+ HBO组、常规治疗+MHT组和常规治疗+HBO+ MHT组,应用原位末端标记法(TUNEL)和流式细胞术对各组大鼠脑挫伤病灶旁的细胞凋亡情况进行检测;使用免疫组化方法检测Bcl-2蛋白在脑组织中的表达情况.结果 常规治疗+ HBO组(16.3±5.0)、常规治疗+MHT组(18.7±6.9)和常规治疗+HBO+ MHT组(24.7±7.2) Bcl-2的表达均高于常规治疗组(8.9±4.2)(P<0.05),其中常规治疗+HBO+MHT组表达最高.结论 HBO联合亚低温治疗脑创伤可有效减少细胞凋亡及增加Bcl-2的表达.Objective To investigate the effects of hyperbaric oxygen (HBO) coupled with mild hypothermia (MHT) on apoptosis and expression of Bcl-2 following traumatic brain injury (TBI).Methods The rat model of traumatic brain injury was developed with the free fall method,then,the animals were randomly assigned to one of the four groups: the routine therapy group,the routine therapy + HBO group,the routine therapy + MHT group,and the routine therapy + HBO + MHT group.Apoptosis of neuron cells adjacent to the traumatic cerebral injury foci was evaluated with TUNEL and flow cytometry,and the expression of Bcl-2 in the brain tissue was detected with immunohistochemistry.Results The expressions,of Bcl-2 in the routine therapy + HBO( 16.3 ±5.0),the routine therapy + MHT( 18.7 ±6.9),and the routine therapy + HBO + MHT groups( 24.7 ± 7.2 ) were all higher than that of the pure routine therapy group ( 8.9 ± 4.2) ( P 〈 0.05 ),with the expression of Bcl-2 in the routine therapy + HBO + MHT group being the highest.Conclusions HBO coupled with MHT could suppress the apoptosis and increase the expression of Bcl-2 efficiently.
