妇科千金片对急性盆腔炎大鼠子宫、卵巢TNF-α、IL-2 mRNA转录水平的影响

摘　　要：目的观察妇科千金片(千斤拔、单面针、金樱根、穿心莲、功劳木、党参、鸡血藤、当归)对急性盆腔炎大鼠子宫、卵巢细胞因子TNF-α、IL-2 mRNA表达的影响,初步探讨该药抗炎免疫作用的分子机制。方法采用混合菌(大肠杆菌、解脲支原体和金黄色葡萄球菌)接种法建立急性盆腔炎大鼠模型。妇科千金片给药后取大鼠子宫、卵巢,提取总RNA,进行RT-PCR反应,通过光密度扫描进行半定量分析。结果与假手术组相比,模型组TNF-α、IL-2 mRNA表达均上升,差异有统计学意义(P<0.05)。与模型组比,妇科千金片治疗组TNF-α、IL-2 mRNA表达均下降(P<0.05)。结论 妇科千金片的抗炎免疫作用可能是通过降低TNF-α、IL-2 mRNA表达,减轻由细胞炎症因子介导的炎症反应来实现的。AIM To investigate the effect of Fuke Qianjin Tablet（Flemingiae prostratae Radix,Zanthoxyi dissiti Radix,Rosae laevigatae Radix,Andrographis Herba,Mahoniae Caulis,Codonopsis Radix,Spatholobi Caulis and Angelicae sinensis Radix） on the expression of cytokines TNF-α and IL-2 mRNA of acute pelvic inflammatory disease in rats,and to explore the molecular mechanism of anti-inflammatory immune of the medicine.METHODSThe rats model for acute pelvic inflammatory disease（Wistar female rats） were successfully made by infecting three various bacteria（Escherichia coli,Ureaplasma urealyticum and Staphyloccocus aureus） into the rats.The rat uteri and ovary were separated and stored after being treated with Fuke Qianjin Tablet,and RNA was isolated and RT-PCR was carried out.The semiquanitative analysis was performed by densitometric scan of agarose gel electrophoresis photos.RESULTS The expression of TNF-α and IL-2 mRNA in the model group overnumbered than simulative control group（P0.05）.After being treated,the expression level of TNF-α and IL-2 mRNA of Fuke Qianjin Tablet group declined（P0.05）.CONCLUSION The possible mechanism of anti-inflammatory immune of Fuke Qianjin Tablet can be regarded as the lowering of the expression of TNF-α and IL-2 mRNA leading to reducing the cell-mediated inflammatory response by the inflammatory cytokines.
