摘 要:为了解北京地区猪瘟病毒(CSFV)流行毒株的遗传变异情况,采用套式RT-PCR方法,对从北京部分地区近期分离的7株CSFV流行毒株的E2基因主要抗原编码区进行了扩增、克隆与序列测定,并应用DNA Star分析软件对所测定的7株毒株与国内外参考毒株的相应序列进行了同源性分析及氨基酸序列比对。结果表明,7株CSFV流行毒株之间核苷酸与氨基酸的同源性分别为96.3%~99.3%和95.6%~100%,与CSFV石门毒株(Shimen株)的核苷酸与氨基酸的同源性分别为81.7%~83.5%和82.4%~84.6%,与CSFV兔化弱毒株(HCLV株)的核苷酸与氨基酸的同源性分别为80.6%~81.7%和78.0%~80.2%,7株CSFV流行毒株均属于基因2群,且705、713、729和734位氨基酸发生置换。本研究初步揭示了北京部分地区目前流行的CSFV毒株与Shimen株和HCLV株在E2基因主要抗原区上存在较大差异。In order to study genetic variation of classical swine fever virus(CSFV)strains in Beijing,the major antigen coding region sequences of E2 gene of seven CSFV strains isolated recently from some areas of Beijing were amplified by nested RT-PCR,cloned and sequenced.The amplified E2 gene sequences of these seven strains were analysed by DNAStar and compared with that of published CSFV strains.The results showed that the homologies of nucleotide and amino acid sequences of the seven strains were 96.3%-99.3%and 95.6%-100% respectively.The homologies of nucleotide and amino acid sequences of the seven strains to Shimen strain were 81.7%-83.5% and 82.4%-84.6% respectively.The homologies of nucleotide and amino acid sequences of the seven strains to HCLV strain were 80.6%-81.7% and 78.0%-80.2% respectively.The seven strains belonged to Group Ⅱ and variations of amino acids were observed at sites 705,713,729 and 734.This study revealed that the major antigen coding region sequences of E2 gene of CSFV strains isolated recently from some areas of Beijing were obviously different from that of Shimen and HCLV strains.
