摘  要:目的探讨地塞米松及1α-25-(OH)2-D3体外诱导致耐受树突状细胞的特性。方法用地塞米松及1α-25-(OH)2-D3诱导CBA/Ca小鼠骨髓来源的树突状细胞,加或不加脂多糖,观察不同树突状细胞的形态学特点,流式细胞仪检测免疫表型,ELISA法检测IL-12p70、IL-10细胞因子分泌,测定不同树突状细胞刺激异基因淋巴细胞增殖能力。结果地塞米松及1α-25-(OH)2-D3诱导的小鼠树突状细胞毛刺少,CD80、CD86水平低,IL-12p70水平降低(P<0.05),IL-10水平升高(P<0.05),刺激异基因淋巴细胞增殖能力受损。结论地塞米松及1α-25-(OH)2-D3体外诱导树突状细胞符合致耐受树突状细胞的特点,为其应用于自身免疫性疾病的治疗提供了理论依据。Objective To investigate the characteristics of dexamethasone and 1α-25-(OH)2-D3 induced dendritic cells.Methods Bone marrow derived dendritic cells were induced with dexamethasone and 1α-25-(OH)2-D3,and treated with or without LPS.Morphology of the dendritic cells was observed.The membrane CD80 and CD86 expression,supernatant IL-12p70 and IL-10 concentrations,stimulating activity for allo-genetic splenic cells were assessed with FACS,ELISA,and MTT,respectively.Results Less and shorter dendrites were observed on dexamethasone and 1α-25-(OH)2-D3 induced tolerogenic dendritic cells,with significantly down-regulated membrane CD80 and CD86,lower supernatant concentrations of IL-12p70 and IL-10(P0.05).Meanwhile,stimulating activity for allo-genetic splenic cells was also impaired in dexamethasone and 1α-25-(OH)2-D3 induced tolerogenic dendritic cells.Conclusion Dexamethasone and 1α-25-(OH)2-D3 induced dendritic cells show tolerogenic characteristics,which provides the evidences for application in treatment of autoimmune diseases.
