板蓝根中总氨基酸含量测定方法的改进

摘　　要：目的改进测定板蓝根中总氨基酸含量的方法。方法以比色法,精氨酸为对照,检测波长568 nm。结果精氨酸在5.32～18.62μg/mL范围内呈良好的线性关系,标准曲线回归方程为Y=50.15 X-0.199 8,r=0.994 7,Y为吸光度,X为浓度。平均回收率100.8%(n=6),RSD=2.98%。结论此法稳定可行,结果准确,专属性好,可用于测定板蓝根中总氨基酸含量。Objective To set up the method for determination of total amino acid in Isatidis radix.Method Colorimetric method was adopted with arginine as control.The detection wavelength is 568 nm.Result There was a good linear relationship in the range of 5.32～18.62 μg/mL for arginine.The standard curve equation was Y = 50.15X-0.199 8,r = 0.994 7,where in Y stands for absorbance and X stands for concentration.The average recovery was 100.8 % with RSD of 2.98 %（n = 6）.Conclusion The method is stable,accurate and specific,and it can be used for the determination of total amino acids in Isatidis radix.
