摘 要:目的研究苦参碱(Matrine,Mat)对大鼠精子膜脂流动性的影响。方法给成年SD雄性大鼠灌服抗生育剂量的Mat(20mg/kg·d-1),6周后,取大鼠附睾精子应用荧光偏振法测定其膜脂流动性,生化法测定精子丙二醛(malondialdehyde,MDA)及超氧化物歧化酶(superoxide dismutase,SOD)含量的变化。结果给药6周后,实验组与对照组比,精子的膜脂流动性降低(P〈0.05),MDA含量增加(P〈0.01),SOD含量减少(P〈0.01)。结论Mat使大鼠精子膜脂流动性降低;精子的氧化应激和有效的抗氧化能力之间的失衡是Mat所致雄性大鼠不育的可能机制。Objective To study the effect of Matrine on the spermatozoa membrane lipid fluidity in male rats. Methods SD male rats were drenched with Mat (20mg/kg ·d) for 6 weeks. The epididymal spermatozoa membrane lipid fluidity was examined by using fluorescence polarization. Biochemical parameters (MDA, SOD) were evaluated with spectrophotometric determination. Results 6 weeks after administration, compared with the control group, MDA level of spermatozoa was significantly higher (P〈0.01); spermatozoa membrane lipid fluidity (P〈0.05) and SOD level (P〈0.01) were lower. Conclusion Matrine can decrease the spermatozoa membrane lipid fluidity in male rats. The imbalance between spermatozoa oxidative stress and the effective antioxidants is a possible mechanism to male rat sterility caused by Mat.
