摘 要:目的研究凉血活血颗粒对急性放射病-肺损伤小鼠模型的保护作用。方法采用5.5 Gy60Coγ射线一次性全身照射小鼠,制成急性放射病-肺损伤动物模型。小鼠在照前45 min至照后30 d,给药组以不同剂量的凉血活血颗粒灌胃给药,正常对照组及模型组用等体积蒸馏水灌胃,均为1次/d。观察照后小鼠的一般情况,照后10 d每组抽取6只检测血常规,照后30 d处死,检测血常规、骨髓DNA、胸腺指数、肺脏指数,用HE染色观察肺脏的病理学改变。结果照后10 d,与正常对照组比较,辐射模型组外周血象显著下降;照后30 d,与正常对照组比较,辐射模型组肺脏指数明显升高,肺脏病理损伤显著,胸腺指数明显降低;外周血象和骨髓DNA有所恢复。凉血活血颗粒高剂量(4 g/kg)组能改善照后30 d小鼠的一般情况,显著降低肺脏指数,减轻辐射造成的肺脏病理损伤,显著增加胸腺指数;改善外周血象和提高骨髓DNA含量。结论凉血活血颗粒对辐射引起的呼吸器官、造血器官和免疫器官损伤具有较好的保护作用。Objective To observe the radioprotective activity of different concentrations of Liangxue Huoxue Granule against whole-body irradiation induced lung injury in mice.Methods A model was established by exposing all the mice to radiation of 5.5 Gy 60Co gamma rays except those in normal control group.45 min before exposure to radiation,the mice began to be administered continuously with different concentrations of Liangxue Huoxue Granule until 30 days after radiation,while in normal and model control group the same amount of distilled water was used once a day.The average life span of irradiated mice was estimated.Meanwhile,peripheral blood parameters,bone marrow DNA,thymus index,lung index and pathological changes of the lung were also observed.Results Compared with non-irradiated controls,irradiation resulted in significantly reducted haematoogical parameters 10 days after irradiation.30 days after irradiation,pathologic injury to the lung was obvious,and thymus index decreased significantly.The peripheral hemogram and bone marrow DNA recovered to some extent.Liangxue Huoxue Granule in high dosage treatment(4 g/kg) group significantly reduced lung index,alleviated pathologic injury to the lung;increased thymus index and improved the peripheral hemogram and bone marrow DNA content.Conclusion Liangxue Huoxue Granule has a better radioprotective activity against whole-body irradiation induced lung injury and against damage to the hematopoietic and immune system in mice.
