摘 要:以不吸水链霉菌武夷变种Streptomyces ahygroscopicus var.wuyiensis为出发菌株,分别用亚硝基胍、氯化锂、微波为单一诱变剂,亚硝基胍和氯化锂为复合诱变剂进行处理,从亚硝基胍处理的菌株中筛选到1株丧失抑菌活性且性状稳定的突变株N1。突变株孢子丝为长螺旋形,孢子呈椭圆形,与出发菌株相比,在形态上没有显著变化且产孢量无变化。突变株的抑菌活性发生了显著变化,对所有8种供试菌株均无抑制作用。HPLC结果显示与出发菌株不同,突变株中抗生素的特征峰消失。本研究为今后开展武夷菌素合成途径和生物合成基因研究奠定基础。Streptomyces ahygroscopicus var.wuyiensis was taken as original strain and its spores were treated by NTG,LiCl,microwave irradiation,complex mutagenesis of NTG and LiCl.One blocked mutant which had no antimicrobial activity named N1 was obtained by NTG treatment.The spore chains were long gyroidal and spores were elliptic.Compared to original strain,the mutant had no significant changes on morphology and sporulation,but lost antibiotic activities against 8 pathogens.HPLC detection indicated that an important component had been lost which was the primary component of wuyiencin.These results make foundation for further research of antibiotic production way and biosynthetic genes of wuyiencin.
