摘 要:目的研究脂质体介导的TK/GCV系统联合普通化疗用于治疗骨肉瘤细胞的可行性,初步研究GCV联合常用化疗药物多柔比星对骨肉瘤mg-63细胞的杀伤作用。方法订购重组质粒pIRES-EGFP-tk,培养和转染骨肉瘤细胞株mg-63,流式细胞仪分析和荧光倒置显微镜下观察细胞生长状态,观察GCV与多柔比星联合应用对mg-63的杀伤作用。结果成功将tk导入mg-63,并能稳定表达。荧光显微镜下观察mg-63(tk+)呈现绿色荧光。GCV(10mg/L)可明显抑制mg-63(tk+)的生长,多柔比星最低有效浓度为(1mg/L);联合应用GCV5mg/L与多柔比星0.25mg/L,即能表现出对mg-63细胞生长明显的抑制作用。结论脂质体介导的TK/GCV可对骨肉瘤细胞产生杀伤作用,与多柔比星联合应用可显著增强多柔比星对转化骨肉瘤细胞的杀伤作用,这将为骨肉瘤的新辅助化疗做出贡献。Objective To investigate the killing effect of TK/GCV system combined with doxorubicin on humanteosarcoma cells and study the efficiency of killing the humanteosarcoma cells by TK/GCV system combined with doxorubicin.Methords Buy the plasmid pIRES-EGFP-tk,and transfect humanteosarcoma cell line mg-63,cell circle analysis and under fluorescence microscopy observe cell growth features,then add the GCV and doxorubicin,study the efficiency of killing the humanteosarcoma cells by TK/GCV system combined with doxorubicin.Results The results showed that TK gene was integrated into mg-63 cells.Observed under fluorescence microscope mg-63(tk+) showing green fluorescence.The growth of mg-63(tk+) cells was greatly inhibited by GCV administration,with 1mg/L as lowest concentration.Doxorubicin killed mg-63 cells at concentration of 1mg/L.When combined with low concentration of GCV(5mg/L) the doxorubicin can be decreased to 0.25mg/L.
