以聚乳酸膜为药物载体研究唑来膦酸局部治疗对去势大鼠骨质疏松骨折愈合的影响

摘　　要：目的研究以聚乳酸膜为唑来膦酸的药物载体局部治疗对去势大鼠骨质疏松骨折愈合的影响。方法选用60只6月龄大SD雌性大鼠,将60只大鼠随机分为3组,每组20只。复制去势骨质疏松大鼠模型并制造左下肢股骨中段横行骨折,后行克氏针髓内固定。A组为空白对照组,B组为实验组(以聚乳酸膜为唑来膦酸药物载体,采用100μg/kg剂量,术中将膜半包覆贴附于骨折断端),C组为实验对照组(以不加载唑来膦酸的膜贴附于骨折断端)。分别于术后1、2、4、6、周行骨密度扫描。结果 A组和B组结果有统计学差异(P<0.05),B组和C组有统计学差异(P<0.05),A组和C组无统计学差异(P>0.05)。结论以聚乳酸膜为唑来膦酸载体局部治疗骨质疏松骨折,对骨折愈合有积极的促进作用;聚乳酸膜对骨折的愈合不产生明显的影响作用。Objective Research with polylactic acid membrane as drug carrier of zoledronic acid in local treatment on fracture healing of ovariectomized rats with osteoporosis.Methods Selection of 60 only 6 months old SD rats.60 rats only randomly divided into 3 groups,each group of 20 only.Replication models in ovariectomized osteoporosis rats run rampant in middle and manufacturing left lower extremity of femur fractures,undergoing intramedullary fixation of Kirschner.A group for blank controlled group,B group for experimental group（to poly lactic acid film for efficacy to phosphine acid drug carrier,used 100μg/kg dose,operation Lieutenant film half package covered posted report Yu fracture broken end）,C group for experimental controlled group（to does not loaded efficacy to phosphine acid of film posted report Yu fracture broken end）.On postoperative respectively 1st,2nd,4th,6th weeks for bone density scans.Results The A group and B group the results statistically difference（P〈0.05）,the B group and C group there were a statistically significant difference（P〈0.05）,the A group and C Group no statistically significant difference（P〉0.05）.Conclusions With polylactic acid membrane as carrier of zoledronic acid in local treatment of osteoporotic fracture,to have a positive role in promoting fracture healing;Polylactic acid membrane does not have a significant effect on fracture healing.
