摘 要:[目的]初步研究紫色链霉菌产褐藻胶裂解酶的最佳培养条件。[方法]以一株具有较高褐藻胶裂解酶活力的紫色链霉菌为出发菌株,考察了底物浓度、氮源、培养温度、培养时间、装液量、KH2PO4浓度、MgSO4.7H2O浓度对产酶的影响。[结果]最佳产酶条件:以蛋白胨和酵母提取物为氮源1,.4%褐藻胶0,.4%KH2PO40,.06%MgSO4.7H2O,培养温度30℃,培养时间6 d。[结论]该研究为紫色链霉菌的发酵产酶及褐藻胶裂解酶的制备奠定了基础。[Objective] The research aimed to study initially the best culture condition of alginate lyase production by Streptomyces violaceoruber.[Method] Streptomyces violaceoruber whose alginate lyase activity was high was as the initial strain.The influences of substrate concentration,nitrogen source,culture temperature,culture time,liquid volume,KH2PO4 concentration,MgSO4·7H2O concentration on the enzyme production were inspected.[Result] The best enzyme production condition: peptone and yeast extract were as the nitrogen source,1.4% alginate,0.4% KH2PO4,0.06% MgSO4·7H2O,the culture temperature was 30 ℃,and the culture time was 6 d.[Conclusion] The research laid the foundation for the fermentation,enzyme production of Streptomyces violaceoruber and the preparation of alginate lyase.
