摘 要:[目的]探讨利用微滤和超滤膜技术分离加工枸杞多糖的最佳工艺条件。[方法]用2540型模设备采取微滤和超滤相结合的试验方法对枸杞多糖浸提液进行分离纯化,结合多糖定量检测结果评价操作压力、操作温度、料液浓度、操作时间等相关因子的膜分离工艺影响因素。[结果]超滤处理的最佳工艺条件为:操作压力0.65 MPa,操作温度45℃,原料液中多糖浓度为0.72 g/L。微滤处理的最佳工艺条件为:操作压力在0.20~0.22 MPa之间,操作温度50℃。[结论]与传统的真空浓缩法相比,膜分离法制备枸杞多糖时能够提高多糖产品的纯度,降低能耗,适合工业化生产。[Objective] To explore the optimum separation process of polysaccharides from FRUCTUS LYCII by microfiltration and ultrafiltration membrane technology.[Method] Polysaccharides were separated and purified from extracts of FRUCTUS LYCII by combining 2540 microfiltration with ultrafiltration membrane.Combined with quantitative detection results of polysaccharides,effects of operating pressure,operating temperature,liquid concentration and operating time on membrane separation of polysaccharides were respectively evaluated.[Result] The optimum conditions of polysaccharides with ultrafiltration membrane were:operating pressure of 0.65 MPa,operating temperature of 45 ℃ and polysaccharide concentration of 0.72 g/L.The optimum conditions of polysaccharides with microfiltration membrane were:operating pressure of 0.2-0.22 MPa and operating temperature of 50 ℃.[Conclusion] Compared with traditional vacuum concentration method,membrane separation for preparing polysaccharides from FRUCTUS LYCII could improve product quality with low energy consuming which was suitable for industrial production of polysaccharides.
