冷冻保存对带血管异体关节移植排斥反应的影响实验研究

摘　　要：目的　观察冷冻保存和环孢霉素A(CyA)对鼠带血管异体关节移植排斥反应的作用。　方法　40只SD大鼠随机分成4组:组、新鲜同系移植;组、新鲜异体移植;组,冷冻保存异体移植;组、冷冻保存异体移植术后用CyA。术后进行血管造影,测定IL-2活性及T细胞亚群(CD4/CD8)的变化,按照Sakai等评分标准进行组织学评分。　结果　组通畅率为0,与其余各组比较差异有显著性(P<0.01)。IL-2及CD4/CD8以组增高最明显,组与组比较差异亦有显著性(P<0.01)。组织学评分:组最低为0.43,组与组比较差异有显著性(P<0.05)。　结论　冷冻保存和小剂量短期应用CyA在抑制鼠异体关节移植排斥方面有协同作用。Objective To observe the effect of cryopreservation and cyclosporine A (CyA) on rejection response to vascularized rat knee joint allografts Methods Employing a model for heterotopic allografts transplantation,40 rats divided into four categories were evaluated Group Ⅰ consisted of SD to SD vascularized knee joint isografts;Group Ⅱ consisted of Wistar to SD vascularized knee joint allografts;Group Ⅲ consisted of Wistar to SD cryopreserved vascularized knee joint allografts,and group Ⅳ consisted of Wistar to SD cryopreserved vascularized knee joint allografts in rats receiving cyclosporine A (5 mg·kg -1 ·d -1 ) The patency of the pedicle was observed through angiography 12 weeks after transplantation Immunologic analysis was performed using IL 2 assay and T lymphocyte subpopulations assay Histological changes of each group were scored according to Sakais criterion Results The patency in group Ⅱ was 0,which statistical difference was significant compared with other groups ( P< 0 01) The changes of IL 2 and CD 4/CD 8 in group Ⅱ were maximal and the difference between group Ⅲ and group Ⅳ reached significance statistically ( P <0 01). The histological score of the grafts in group Ⅱ was the lowest (0 43) There was significant difference statistically in histological score between group Ⅲ and group Ⅳ (0 01< P <0 05) Conclusions Cryopreservation combined with low dose and short term CyA has a synergistic effect on rejection response to vascularized rat knee joint allografts
