摘  要:目的探讨阿魏酸衍生物对心肌细胞缺氧复氧(H/R)损伤的影响。方法用1~3d的SD大鼠进行心肌细胞原代培养,取生长5~6d的心肌细胞分为对照组、H/R组、溶剂组、不同阿魏酸衍生物组,除对照组外其他组均做H/R模型。通过测定肌酸激酶(CK)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)及丙二醛(MDA)的变化,观察不同阿魏酸衍生物对心肌细胞H/R损伤的保护作用。结果与H/R比较,除IIa外,阿魏酸类似物均可明显减少心肌细胞H/R损伤所造成的CK、LDH的渗出、MDA含量,同时可增加SOD酶活力。结论阿魏酸衍生物苯环上3,5位取代基可以明显影响此类化合物抗氧化活性,甲氧基和乙氧基表现为提高活性,溴次之。Objective To study the effects of ferulic ccid analogs in a cardiomyocyte Hypoxia-reoxygenation(H/R)model.Methods The cardiomyocyte from neonatal SD rats were isolated and cultured.The H/R models of neonatal rat cardiomyocyte were established.The cardiomyocyte were randomly divided into control group,H/R group,solvent group,and Ferulic Acid analogs group.Measurements included cell damage evaluated by Lactic Acid Dehydrogenase(LDH)and Creatine Kinase(CK)release in the medium were measured,the contents of intracelluar Malonaldehyde(MDA),the activity of Superoxide Dismutase(SOD)of the cultured myocardial cell were measured at the end of experiment.Results Except for IIa,FA and some its analogs significantly attenuated levels of LDH,CK,MDA and increased activity of SOD.Conclusion 3 and 5 substituted group on benzene ring of Ferulic acid and its analogs could obviously influence the antioxidant activities,that could be increased by methoxyl,oxethyl or bromine on benzene ring.
